Devices and methods are disclosed herein for a biomimetic flow apparatus. The biomimetic flow apparatus includes a microfluidic flow channel and an inlet in fluid connection with the flow channel for allowing fluid to flow into the flow channel. The flow channel has at least one surface with a topography formed therein. The topography of the at least one surface of the flow channel is selected to cause cells in a cell layer above the surface to achieve an arrangement, behavior, or morphology. The cell arrangement, behavior, or morphology is determined at least in part by the topography of the at least one surface.

This invention describes a cell growth apparatus, particularly neuronal printed circuit board apparatus comprising an aerogel base and a pre-printed cellular growth pattern. The cellular growth pattern is comprised of combinations of layers of cellular adhesion promoting materials, cellular adhesion inhibiting materials, and/or cellular signal promoting materials. The invention further describes methods of promoting cell growth using the neuronal printed circuit board apparatus of the invention. The invention is useful for regeneration and precise guidance of cells, particularly nerve cells, when used as an implant.

Composite structures and methods for generating micropatterned materials suitable for use in cell culture applications are disclosed. The improvement of these compositions and methods over the prior art is based on the unexpected discovery that minor chemical modifications can be introduced to greatly enhance the adherence and/or stability of a cell- adhesive material. The micropatterned materials are inexpensive to manufacture, have long shelf- life, and are stable for prolonged periods of time under cell-culture conditions. Moreover, biologists can use these micropatterned substrates with the same ease as conventional cultureware and without the need for special sample preparation.

Use of this technical means makes it possible to culture cells originating in a specific single cell, to culture and observe the cells while specifying cells under interaction during the culture, to observe a difference between a specific cell exclusively to which a chemical (a signal, etc.) has been sprayed, and other cells having been cultured while maintaining the cell density at a constant level, and the like. A novel means whereby cells in a specific state can be exclusively collected and a gene in the cells, an mRNA expressed therein, etc. can be analyzed or biochemically assayed.

A system is provided for selective clonogenic expansion of relatively undifferentiated cells, including (a) a tube containing a plurality of beads of a size which permits a plurality of the undifferentiated cells to grow thereon (Figure 1), the beads bearing on their surfaces a selective binding molecule which binds to a surface antigen present on the relatively undifferentiated cells (Figure 1A), wherein the antigen is not present on the surfaces of the relatively differentiated cells; (b) means for continuously providing nutrients to the relatively undifferentiated cells growing on the beads, wherein the nutrients are delivered via a fluid which flows through the tube and past the beads (Figure 1); and (c) means for continuously harvesting the relatively undifferentiated cells downstream of the beads (Figure 1).
