Abstract:
A method of identifying potentially useful treatments which restore the function of CFTR or other related protein mutations is disclosed. The method involves creation of yeast STE6-CFTR chimeric sequences which upon introduction of a desired mutation inhibit expression of the yeast STE6 gene. A yeast assay (STE6 DELTA ) transformed to include the hybrid chimeras is then exposed to a potential treatment for the mutation. A treatment which restores the function of the STE6 gene and thereby allows mating of the yeast strain is indicative of a potential remedial treatment for the mutation in this selected protein.
Abstract:
The electrophoretic isolation and monosaccharide sequence determination of a neutral or weakly acidic oligosaccharide species of interest are disclosed. A labeling compound and a charged group are coupled to the reducing end of the species of interest, thereby facilitating electrophoretic separation and detection of the separated species. The resolved species of interest can then be recovered from the electrophoretic medium, for example, by electrophoretic transfer to a charged solid support. Following isolation, monosaccharide units can be cleaved successively from the non-reducing end of the species of interest to reveal the monosaccharide sequence. The identity of each monosaccharide unit is determined by correlating cleavage data with known exoglycosidase specificites.

Analogs are prepared of the carbonic anhydrase inhibitors methazolamide, and dichlorphenamide. The analogs link a cleavable functional group to the pharmacologically active carbonic anhydrase inhibitor resulting in compounds of improved corneal permeability, which are non-irritable to the eye and which are effective topically to inhibit intraocular pressure.

The invention pertains to the dystrophin-glycoprotein complex of mammalian skeletal muscle and a method of isolating said complex. The components of the complex and methods of separating and isolating said components also pertain to the invention. In addition, the invention further relates to a method of detecting and quantifying the loss of a non-dystrophin component of the dystrophin-glycoprotein complex with said loss being indicative of muscular dystrophy.

A method of making a sugar-based polymer is disclosed. A sugar and an organic acid derivative having at least two carboxylate functionalities are mixed in a substantially non-aqueous organic solvent with a hydrolytic enzyme and, thereafter, the resulting mixture is agitated for a time sufficient to allow for the polymerization of the sugar. The resulting sugar-based polymer has sugar incorporated in the polymer backbone.
