Described herein are multi-purpose substrates composed of (1) a base coated with a calcium phosphate coating and (2) a fluorophore-labeled collagen adsorbed on the calcium phosphate coating. The multi-purpose substrates are useful in culturing and studying the activity of a variety of cells. The multi-purpose substrates described herein can be used for both solution- and image -based analysis of cultured cells. New methods for producing and using such coated substrates are also disclosed.

An ex vivo method of producing osteoclasts is described that includes providing isolated CD 11 C + dendritic cells and culturing the CD 11 C + dendritic cells in culture medium under conditions effective to produce osteoclasts. Also disclosed are methods of up-regulating or down-regulating bone resorption by manipulating the osteoclastogenesis of CDl Ic + dendritic cells either in vivo or in vitro. Methods of treating an inflammatory bone disease or a metabolic bone disorder in a subject, and screening assays to identify compounds or genes that affect myeloid osteoclastogenesis are also described.

The invention relates to the use of in vitro isolated or in vitro differentiated osteoclasts for bone rebuilding, in particular by in vivo application, to trigger bone rebuilding by attracting osteoblasts and preferably also vascular cells to sites of bone defect. It has been found by the inventors that osteoclasts can control bone rebuilding by secreting chemotactic factors and attracting osteoblasts and vascular cells to a site of bone defect. A method of inhibiting osteoclast formation including inhibiting eosinophil chemotactic factor-L expression or activity. This may be accomplished by a number of means, including the use of anti-ECF-L antibody, antisense S-oligonucleotide to ECF-L, mECF-L polyclonal antisera, rabbit preimmune antisera, OPG RANK-Fc and combinations thereof, as well as other inhibiting materials. In another embodiment, osteoclast formation is inhibited by inhibiting RANKL formation. In a further embodiment, a method of inhibiting osteoclast formation is accomplished by means of mECF-L in the presence of RANKL.
