A system for seeding cells onto an array of screens or scaffolds is provided. The system can include a storage screen holder and a loadable screen holder, which can be in spaced relationship such that screens can move from the storage screen holder into the loadable screen holder, when urged by a pusher mechanism. Cells for seeding can be dispensed by a dispenser. A motion control system can move appropriate ones of the screens, screen holders and dispenser as needed. The lowest screen can be moved into the loadable screen holder, and cells can be dispensed onto it, and then successively higher screens can be moved into the loadable screen holder and seeded with cells. Components such as the storage screen holder the loadable screen holder and the screens can be supplied as a pre-sterilized assembly.

There is an urgent need for new antibiotics and antimicrobials to treat and cure acute and chronic infections by bacterial pathogens. Current high-throughput practices, do not account for any specifics in bacterial growth or inhibition. Such approaches inefficiently identify many false positives and false negatives of novel compounds. Additionally, this strategy informs little about biochemical changes resulting from bactericidal exposure. We disclose a Precision Throughput approach that can be used to effectively screen new potential antimicrobials under specialized growth conditions relevant to bacterial pathogenesis. This platform can be used to determine new biology and antimicrobial effects of the pathogens Pseudomonas aeruginosa and Group A Streptoccoccus and determine new metrics for characterizing bactericidal activity by a Precision Throughput approach.

A bona fide adaptable in vitro microcirculation model is provided by integrating a 3-D printed network of endothelial-cell lined perfusion channels, formed via sacrificial casting in a gel matrix, with a native, adaptable microvasculature matured from native microvessels added to the gel matrix. Responsive vascular adaptation exhibited by the in vitro microcirculation is physiologically relevant. Methods for fabricating, devices, models and investigative platforms for pharmaceutical applications, vascular mechanism and microvessel-parenchyma interaction studies, and vascularizing strategies for tissue engineering applications are also disclosed.

The invention features devices and kits for capturing and culturing microorganisms (e.g., bacteria, fungi, or protists) and methods of using the devices and kits to detect microorganisms in environmental and other samples. The device includes a nutrient media having a flat growth area on which microorganisms can grow. Samples are collected by contacting the device with any environmental sample, e.g., rolling device on a work surface or exposing device to air, or by filtering a sample through a membrane. Microorganisms deposited on the membrane derive nutrients from the underlying media and grow into colonies that can then be detected using methods known in the art. The detected colonies can be imaged digitally or with film.
