The invention relates to a method for determining the quantity of pre-synaptic neuromuscular blocking substance (notably botulinum toxin) contained in a sample. In one aspects, the method comprises the following steps: (i) determining the minimum voltage Vm needed to induce the contraction of muscle tissue, said muscle tissue being connected to an electrical stimulator through a motor nerve and preferably immersed in an oxygenated physiological buffer containing glucose; (ii) adding the sample containing the pre-synaptic neuromuscular blocking substance; (iii) electrically stimulating, at a voltage at least equal to Vm, the muscle tissue at certain time intervals; (iv) comparing the effect induced by the sample to the effect induced by a reference substance and thereby determining the quantity of the pre-synaptic neuromuscular blocking substance in the sample.

Monkey alpha-7 neuronal nicotinic acetylcholine receptor polypeptides, as well as the DNA (RNA) encoding such polypeptides, are disclosed. Also disclosed are methods for utilizing such polypeptides in diagnostic assays for identifying mutations in nucleic acid sequences encoding the polypeptides of the present invention, for detecting altered levels of the polypeptide of the present invention as a means of detecting diseases and methods of screening potential modulators of the novel alpha-7 receptor disclosed herein. Transgenic animals expressing polypeptides disclosed herein are also described.

The present invention provides novel methods and diagnostic kits for the objective measurement of the severity of pain or stress being experienced by a patient with a disorder, diagnosis and treatment for patients suffering from painful disorders, and monitoring the effectiveness of different pain-treatment protocols. Pain-measuring methods comprise collecting a sample from a patient and determining the presence of a pain-associated marker in the sample. Methods for alleviating pain comprise administrating a dose of a therapeutically effective amount of a composition to the patient wherein the dose is determined by the presence of a pain-associated marker in a biological sample obtained from the patient. Compositions for alleviating pain comprise substances that are pain-associated markers or agents that interfere with pain-associated markers, and block or modulate the progression of pain perceived by the patient.
