Méthode d'analyse quantitative d'un mélange de composés moléculaires par chromatographie en phase gazeuse bidimensionnelle. On réalise une chromatographie en phase gazeuse bidimensionnelle au cours de laquelle on enregistre un signal chromatographique. De ce signal, on génère un chromatogramme en deux dimensions sur lequel des pics chromatographiques forment des taches. On délimite ces taches à l'aide de polygones. Puis, pour chaque polygone, on extrait des portions du signal chromatographique comprises entre deux intersections de ce polygone avec des colonnes du chromatogramme. On détermine des temps de début et de fin de pics chromatographiques présents sur ces portions, et l'on ajuste le polygone en déplaçant ses points d'intersection en fonction des temps de début et de fin de pics chromatographiques. Enfin, on détermine des quantités de composés moléculaires en calculant l'aire des polygones ainsi ajustés. Application notamment à l'analyse quantitative de produits issus de l'industrie chimique ou pétrolière.

The present invention provides a chromatograph/mass spectrometer capable of producing a composite chromatogram from plural chromatogram signals without causing the baseline level to rise or the intensity of the chromatogram peak to decrease. The chromatograph/mass spectrometer includes a chromatograph unit for separating a sample into components in the order of time, a mass spectrometer unit for analyzing the components of the sample, and a signal generator for producing a signal carrying a chromatogram obtained by the analysis. It also includes a chromatogram composer for receiving signals of plural chromatograms simultaneously measured under different conditions, for choosing the largest value of the plural signals for each point in time, and for integrating the chosen values to produce a composite chromatogram.

The method and apparatus effects real time identification and determination of the boundaries of these peaks for the purpose of fraction collection. The method and apparatus determines the boundary of a peak within a detector response profile that comprises data points plotted graphically on an X and Y-axis wherein the X-axis represents a first variable and the Y-axis represents a second variable, each variable having a value. The plot at each of the data points has a slope. Data points having a slope which deviates from a consistent value define a peak, and data points having a slope with a consistent value define a baseline.

A method (200) for performing separation assays of biochemical samples includes computing a quality metric (210) based on peak data produced during the separation run. The quality metric is the basis for selecting a subsequent step in the assay, including whether to re-run the separation when the quality metric indicates a low quality separation run. In a preferred embodiment, the quality metric is computed based on a peak resolution metric indicative of the peak resolution of the sample peaks in the data and a signal-to-noise ratio of the data. When a co-migrating standard is included in the separation run, the quality metric is further based on the degree of migration linearity of the reference peaks produced by the standard. The method was reduced to practice in separations to size and sort DNA fragments in high-throughput capillary array electrophoresis separations.

The invention concerns a method for the realignment of peaks in gas chromatographic analyses after the replacement of the used capillary column with a nominally equal one, wherein a K coefficient characteristic of the column is preventively calculated by flowing carrier gas in the column under stabilized conditions of pressure and flow rate and of the column temperature, said K coefficient being stored in a memory of the gas chromatographic system. Said K coefficient is used to correlate at least two of the above mentioned parameters in subsequent analyses, obtaining an automatic realignment of peaks. The invention also concerns the analytical equipment to perform the method, and the capillary gas chromatographic column characterized as reported above (fig. 1).

In a chromatograph comprising a pump, a sample injecting unit, a column, a photo-diode array detector and a data processing unit having a spectrum library of registered spectrums of known components, a spectrum selected in advance from the spectrum library is compared with a spectrum of each of peaks in an actual sample. Thereby, it is possible to reduce number of spectrum comparisons required for identification, and it is further possible to perform identification speedy since the identification can be performed during analyzing.

In a two-dimensional gas chromatography system (200), it is necessary to "pair" peaks (1A1, 1A2, 1A3, 1A4, 1A5) from one chromatogram with respective peaks (1B1, 1B2, 1B3, 1B4, 1B5, 1B6) of another chromatogram. Both peaks of a pair should correspond to the same sample component. Confirmation or disconfirmation of pairs may be speculative or arbitrary. A hybrid chromatographic column (2AB) is designed so that the retention time of a sample component is the average of the retention times of that component in the two independent columns (2AA, 2BB). Thus, a peak location in the hybrid chromatogram can be calculated for each pair of peaks. The absence of a peak at that location or the inconsistency of the area of a peak at that location disconfirms the pairing. Higher dimensional systems and other separation technologies may also be used.
