An automatic apparatus for use in assays for separating magnetic particles includes means for separating magnetically responsive particles from a liquid dispersion disposed in a plurality of reaction vessels, and transporting means for transporting the reaction vessels in sequence past at least one processing position. A robotics reagent arm and probe dispense reagents into the reaction vessels and a reaction monitoring device is capable of relative movement with respect to the transporting means. Incomplete separation is effected by positioning a magnetic field in contact with the reaction vessel for a first shortened time interval during which the particles partially aggregate and afterwards removed from the reaction vessel. The magnet is repositioned in contact with the reaction vessel for a third time interval to achieve full separation of particles from the liquid.

A method of quantitative analysis of cholesterol contained in a high-specific-gravity lipoprotein by adding to a lipoprotein-containing specimen a substance which forms a complex with a lipoprotein other than the high-specific-gravity lipoprotein and a surfactant, and enzymatically determining the cholesterol content. The method permits the quantitative analysis of cholesterol contained in a high-specific-gravity lipoprotein with a simple operation at a high efficiency without the necessity for pre-treatment such as centrifugal separation. This method is applicable to various automatic analyzers and useful also in the field of clinical examination.

The measurement of imipramine or desipramine is accomplished in a specific immunoassay employing antibodies prepared with imipramine or desipramine derivatives of formula (III), wherein P is an immunogenic carrier material, X is two heteroatoms, Y is a linking group comprising from 1 to 6 carbon atoms and P is an immunogenic carrier material, and wherein for imipramine, R is CH3, and for desipramine, R is H. The present invention also describes the synthesis of unique labeled reagents of the structure of formula (IV), wherein Z is a linking group comprising 1 to 4 carbon atoms and 0 to 2 heteroatoms and Q is a detectable moiety, preferably fluorescein or a fluorescein derivative, and wherein for imipramine, R1 is CH3, and for desipramine, R1 is H.

A method and device for testing for the presence of substances such as drugs in body fluids while simultaneously positively identifying the test subject. The device comprises an absorbent pad and a membrane mounted to the absorbent pad containing a plurality of separated areas provided with different immobilized ligand having a specific receptor site for capture or rejection of specific antigens produced by different predetermined drugs. The method of testing comprises the steps of: (a) obtaining a quantity of body fluid in a container, (b) adding an antibody to the body fluid to form a mixed solution, (c) placing the mixed solution on a membrane and absorbent pad device and immobilized ligands, (d) covering the finger with a thin film of labelled antibody and (e) placing the finger on the membrane containing immobilized ligand specifically selected to bind to labelled antibody presenting the fingerprint of the test subject, the presence or absense of substances being tested for in the body fluid so that solution is deposited from the finger on the membrane indicating presence or absence of the substance.
