The present invention relates, in general, to attenuated swine influenza viruses having an impaired ability to antagonize the cellular interferon (IFN) response, and the use of such attenuated viruses in vaccine and pharmaceutical formulations. In particular, the invention relates to attenuated swine influenza viruses having modifications to a swine NS1 gene that diminish or eliminate the ability of the NS 1 gene product to antagonize the cellular IFN response. These viruses replicate in vivo, but demonstrate decreased replication, virulence and increased attenuation, and therefore are well suited for use in live virus vaccines, and pharmaceutical formulations.

This invention provides a genetically engineered attenuated swine influenza virus, wherein the virus comprises a swine influenza virus NS1 gene with a mutation that results in a swine influenza virus NS1 protein carrying a deletion within its carboxy-terminus. The invention also provides a composition comprising said genetically engineered attenuated swine influenza virus. Additionally, a genetically engineered swine influenza virus is provided for immunization of a pig or prevention of a disease or symptom associated with swine influenza virus. The invention also provides a cultured cell or an embryonated egg containing said genetically engineered attenuated swine influenza virus and a method for production of an immunogenic composition or a pharmaceutical composition.

This invention provides an attenuated virus which comprises a modified viral genome containing nucleotide substitutions engineered in multiple locations in the genome, wherein the substitutions introduce synonymous deoptimized codons into the genome. The instant attenuated virus may be used in a vaccine composition for inducing a protective immune response in a subject. The invention also provides a method of synthesizing the instant attenuated virus. Further, this invention further provides a method for preventing a subject from becoming afflicted with a virus-associated disease comprising administering to the subject a prophylactically effective dose of a vaccine composition comprising the instant attenuated virus.

The present invention relates to an advantageous process for industrially producing an inactivated virus (e.g., HVJ, etc.) envelope. To solve the problem, the present invention provides a process for treating a virus with an alkylating agent to produce an inactivated virus envelope. A vector for introducing a biological macromolecule, such as a gene or the like, which is prepared from the inactivated HVJ envelope of the present invention, can be used for genetic function analysis or gene therapy. Specifically, the present invention comprises the steps of (a) inactivating a virus with an alkylating agent, (b) obtaining a condensate solution of the virus or the inactivated virus, and (c) purifying the virus or the inactivated virus by column chromatography and then ultrafiltration. The present invention also provides a composition and pharmaceutical agent which utilize an envelope obtained by the process of the present invention.
