In several embodiments, the MPV F proteins and/or nucleic acid molecules can be used to generate an immune response to MPV in a subject. In additional embodiments, the therapeutically effective amount of the MPV F ectodomain trimers and/or nucleic acid molecules can be administered to a subject in a method of treating or preventing MPV infection.

The present invention provides an isolated mammalian negative strand RNA virus, metapneumovirus (MPV), within the sub-family Pneumoviridae, of the family Paramyxoviridae. The invention also provides isolated mammalian negative strand RNA viruses identifiable as phylogenetically corresponding or relating to the genus Metapneumovirus and components thereof. In particular the invention provides a mammalian MPV, subgroups and variants thereof. The invention relates to genomic nucleotide sequences of different isolates of mammalian metapneumoviruses, in particular human metapneumoviruses. The invention relates to the use of the sequence information of different isolates of mammalian metapneumoviruses for diagnostic and therapeutic methods. The present invention relates to nucleotide sequences encoding the genome of a metapneumovirus or a portion thereof, including both mammalian and avian metapneumovirus. The invention further encompasses chimeric or recombinant viruses encoded by said nucleotide sequences. The invention also relates to chimeric and recombinant mammalian MPV that comprise one or more non-native or heterologous sequences. The invention further relates to vaccine formulations comprising mammalian or avian metapneumovirus, including recombinant and chimeric forms of said viruses. The vaccine preparations of the invention encompass multivalent vaccines, including bivalent and trivalent vaccine preparations. The invention also provide methods for propagating virus.

In particular, the heterologous gene products include gene product of another species of PIV or from another negative strand RNA virus, including but not limited to, influenza virus, respiratory syncytial virus, human metapneumovirus and avian pneumovirus. The chimeric viruses and expression products may advantageously be used in vaccine formulations including vaccines against a broad range of pathogens and antigens.

The invention aims at improving the efficacy and protection induced by DNA vaccination against viruses of the family of Paramyxoviridae and against the herpes virus, in pets and sport animals. The improvement of DNA vaccination is achieved either by formulating the vaccine with a cationic lipid containing a quaternary ammonium salt, DMRIE, or by modifications in the nucleotide sequence coding for the antigen of interest in particular of deletions of the fragment of the nucleotide sequence coding for the transmembrane domain of the antigen of interest, and/or insertions of introns and/or insertions of nucleotide sequences coding for the signal peptides, or by adding GM-CSF, or by combinations thereof. The invention also concerns the resulting vaccines.

An avian vaccine formula including at least three polynucleotide vaccine valencies that each include a plasmid containing an avian pathogen valency gene capable of being expressed in vivo in host cells. Said valencies are selected from the group which consists of Marek's disease virus, Newcastle disease virus, infectious bursal disease virus, infectious bronchitis virus and infectious anaemia virus. The plasmids include one or more genes per valency, and said genes are selected from the group which consists of gB and gD for Marek's disease virus, HN and F for Newcastle disease virus, VP2 for infectious bursal disease virus, S, M and N for infectious bronchitis virus and C + NS1 for infectious anaemia virus.
