Cellulase may be prepared in good yield and at relatively low cost by culturing certain mutant strains of the genus Trichoderma, which exhibit increased inducibility of cellulase by L-sorbose, in medium wherein the carbon source comprises cellulose-containing material of plant origin, for example, bagasses, waste papers, rice plant hulls and straws or soybean wastes. Preferred mutant strains are exemplified by Trichoderma reesei PC-1-4, PC-3-7, X-30 and X-31.

A process for producing enzymes capable of catalyzing the hydrolysis of cellulose comprising the steps of (a) inoculating a growth medium containing an inducer of cellulose synthesis with T. reesei strain MCG 80 and (b) incubating the inoculated growth medium under conditions which will permit the growth of MCG 80. The novel biologically pure MCG 80 strain of T. reesei is derived from T. reesei strain RUT-C 30.

DNA fragments containing an alkaline cellulase K gene, which comprise about 4.0, about 2.4 and about l.9 kilo base pairs, respectively, each having a specific restriction map, recombinant plasmids containing any of these DNA fragments, and recombinant microorganisms harboring any of said plasmids. The alkaline cellulase K gene is derived from a bacterial strain which belongs to the genus Bacillus and is alkalophilic, that is, capable of revealing an optimum growth in an alkaline pH region.

A vector system for transformation of Trichoderma has been developed. The vector system can be used for high expression and secretion of proteins in Trichoderma by transformation of a suitable Trichoderma strain with the vector system comprising a gene for a desired protein.
