Novel DNA sequences which are responsive to the presence of methanol, catabolite non-repressing carbon sources and carbon source starvation are provided. In addition, novel constructs including these DNA sequences, as well as transformed organisms therewith are provided. Processes for producing the DNA sequences and constructs of the invention are detailed. The production of polypeptide product under the control of the regulatory regions of the invention is demonstrated.

The present invention relates to a stable, allopolyploid, somatic fusion product useful in the fermentation arts and especially in the production of fuel alcohols. More particularly, the present invention relates to a novel allopolyploid yeast strain, Saccharomyces diastaticus NCYC 1460, which is the product of the spheroplast fusion of: (a) a hybrid, diploid strain of Sacch.diastaticus; and (b) a fusion partner which is a polyploid brewing yeast strain of the species Saccharomyces uvarum (carlsber- gensisJ. The fusion product of the present invention is capable of an enhanced rate of fermentation and a generally higher degree of final attenuation than is typical of either of the fusion partners. Moreover the fusion product also evidences enhanced osmo- and thermo-tolerances and its multiple, non-allelic DEX gene complement renders the organism well suited to the fermentation of substrates containing dextrins and oligosaccharides.

An improved process for producing an organoiron (II) compound-containing antianaemic composition which comprises subjecting a yeast to anaerobic culturing in a saccharide-containing nutrient medium therefor in the presence of an iron compound. By the process, an antianaemic composition which is very stable and excellent in absorbabil- i ty into a living body and incorporation of iron into hemoglobin can be produced in high yield in a shortened period of time.

A gene having a DNA sequence complementary to that of the glucoamylase polypeptide mRNA from a fungal species, preferably Aspergillus awamori, is prepared. The mRNA is an approximately 2.2 kilobase poly a RNA obtained from fungal cells grown under conditions of glucoamylase induction. Reverse transcription of the mRNA provides a glucoamylase probe used to identify genomic digest fragments containing glucoamylase gene regions, which are sequenced to locate the introns and exons. The genomic fragments are spliced together to form a gene having a DNA sequence with altered or deleted introns which codes for fungal glucoamylase protein and is capable, when correctly combined with a cleaved DNA expression vector, of expressing a non-native protein having glucoamylase enzyme activity upon transformation of a host organism by the vector. The host organism is preferably a bacterium or a yeast. The transformed host organism may be used to produce ethanol.
