    摘要: Cytotoxic T lymphocytes (CTLs) specific for antigenic peptides derived from IgE molecule can be generated in vitro by stimulating resting naive CD8 T cells with IgE peptides presented by artificial antigen presenting cells. The IgE specific CTLs lyse the target cells loaded with IgE peptides in vitro and inhibit antigen specific IgE response in vivo. In addition, adoptive transfer of the IgE specific CTL to an asthmatic mouse model can inhibit the development of lung inflammation and airway hypersensitivity. IgE specific CTL provides a treatment for allergic asthma and other IgE-mediated allergic diseases. Antigenic peptides identified from non-tumor self-antigens induce specific cytotoxic T lymphocyte (CTL) in vitro. The CTL induced by peptides identified from CD40L can kill activated CD4 T cells. In vitro generated CTL specific for CD40L inhibit CD4-dependent antibody responses of all isotypes in vivo. In contrast, CTL induced by antigenic peptides derived from IgE specifically inhibit IgE responses, and adoptive transfer of CD40L-specific CTL to NOD mice at early age delay the development of diabetes in NOD mice. In vitro generated CTL specific for non-tumor self-antigens expressed on activated CD4 T cells regulate immune responses in vivo.

    摘要: The present invention provides reactive aqueous or solvent borne compositions comprising one or more low-temperature curing alkylether functional aminoplast resins having an average degree of polymerization of from 1.0 to 2.0, preferably alkylether group containing melamines, one or more hydroxyl group containing resins or polymers, such as acrylics and polyesters, one or more unblocked acid catalysts each having a pKa of 0.25 to 1.0, such as organic sulfonic acids, and one or more volatile amines, e.g. N-dipropylamine, that have a relative evaporation rate (RER) of 50-500, wherein, RER equals (p)(M)/11.6, with p being the vapor pressure of the amine in mm Hg and M being the molecular weight of the amine. The compositions cure at from 60° C. to 135° C., e.g. below 105° C., to form durable coatings suitable for plastic substrates, such as automotive plastics. The present invention also provides methods of forming the compositions comprising mixing the said aminoplast resins, hydroxyl group containing resins or polymers, unblocked acids and volatile amines to form a mixture, wherein the acids and the amines are added separately into the mixture.

    摘要: A new method to capture, purify and expand antigen-specific T lymphocytes has been developed using magnetic beads coated with recombinant MHC class I molecules. This method was optimized using homogenous populations of naive T cells purified from mice transgenic for the 2C T cell receptor (TCR). These T cells were captured on beads coated with MHC class I molecules and the relevant antigenic peptides. MHC and peptide specificity was confirmed by the usage of irrelevant MHC peptide combinations. An enrichment of 800 to 1600 fold was measured, using 2C T cells mixed with irrelevant T cells, starting from a 2C T cell frequency of 1/3000. The same approach was used to purify antigen-specific CD8+ T cells from total CD8+ T cells from naive mice. The recovered cells could be expanded and specifically kill target cells in vitro; they had a significant effect in vivo as well. We expect this procedure to be suitable to purify and expand in vitro tumor- and virus-specific killer T cells for use in cell therapy.

A z-lens upstream of the analyser can be switched between a high sensitivity mode wherein a beam of ions passing therethrough is substantially focused on to the entrance slit of the analyser, and a low sensitivity mode wherein the beam of ions is defocused so that the diameter of the beam substantially exceeds that of the entrance slit of the analyser. Obtaining data in the low sensitivity mode in combination with obtaining data in the high sensitivity mode enable an order of magnitude increase in the dynamic range to be obtained.
