摘要： A multilayer analysis film for assaying transaminase in a coupled reaction system by a kinetic method comprises (1) a porous layer containing a transaminase substrate, (2) a coloring indicator layer for the detection of hydrogen peroxide, and (3) a transparent support, integrally laminated in this order. Pyruvate oxidase is located in a large excess in the uppermost porous layer where a rate-determining step receives maximum exposure to oxygen in the air. By the use of the analysis film, transaminase activity can be assayed with high sensitivity.

摘要： An analysis film for assaying transaminase activity comprises a reagent layer containing pyruvic acid oxidase, a combination of a hydrogen donor and a coupler--which is capable of forming a cationic dye--and an anionic polymer. The cationic dye formed by the reaction with a color indicator composition for detecting hydrogen peroxide is fixed with the anionic polymer, thereby mobilization and diffusion of the cationic dye being prevented so that the analysis film achieves high detection sensitivity and high accuracy.

摘要： A quantitative analysis film comprising a reagent layer comprising a cationic dye-forming color indicator for detecting hydrogen peroxide and an anionic polymer. By the use of such a color indicator and an anionic polymer in combination, a cationic dye is formed; the formed cationic dye is then bound and fixed to the anionic polymer to provide an intensified color. A method for the colorimetric assay of hydrogen peroxide or a precursor thereof with high sensitivity and high efficiency is also disclosed.

The enzyme has an optimum pH in a in pH range within which the dissociation of p-nitrophenol is insufficient for color formation. The layers of the element comprise a self-developing substrate having p-nitrophenol group attached to the molecular structure through an ether or ester linkage and a cationic polymer, said self-developing substrate and said cationic polymer being incorporated separately in different water-retaining carriers which are arranged to allow the layers to have liquid contact with each other.
