Provided is a cell culture apparatus for culturing cells, and optionally, for performing magnetic separation of cells desired to be cultured. The cell culture apparatus preferably comprises a frame; two membranes, preferably each being gas permeable, which are securedly sealed in a leak-proof selaing to a frame in forming a culture chamber therebetween; and at least one resealable aperture to allow substances to be introduced into, or withdrawn from, the culture chamber.
Abstract:
Compositions are provided for detecting Tn antigen expressing-shed tumor antigen. The compositions include peptides having binding specificity for the Tn antigen; and a detector molecule comprising a peptide, according to the present invention, which is bound to a detectable label. The compositions may be used in a method to detect lymphoid tissues containing shed tumor antigen expressing the Tn antigen.

Provided are methods for purifying functionalized fluorescent nanocrystals having affinity ligand operably bound thereto by using a solid support matrix in a reactor through which solutions are circulated, and by using an immobilized solid phase formed by operably binding functionalized fluorescent nanocrystals to the solid support matrix. Also provided are purified functionalized fluorescent nanocrystals having affinity ligand operably bound thereto which are purified using the methods according to the present invention to be substantially free of free affinity ligand.

Provided are a fluorescent microsphere comprised of a plurality of fluorescent nanocrystals operably bound to a polymeric microsphere, and a method of producing the fluorescent microspheres which comprises contacting the polymeric microsphere with a plurality of fluorescent nanocrystals under suitable conditions in which the fluorescent naocrystals become operably bound to the polymeric microsphere. Also provided is a method of using the fluorescent microspheres capable of determining the presence or absence of a predetermined number of analytes in a sample by contacting the sample with the fluorescent microspheres, and detecting the fluorescence signal pattern of excited fluorescent microspheres bound to one or more analytes of the predetermined number of analytes, if present in the sample.

A method for screening for a pro-tumor immune response according to the present invention, the method comprises: contacting a clinical sample with one or more detector molecules for detecting, and then determining the amount of, mononuclear cell phenotype in the sample; and (b) comparing the amount of mononuclear cell phenotype determined in the sample to a reference value for the mononuclear cell phenotype; wherein a significant difference in the amount of mononuclear cell phenotype determined as compared to the reference value may be an indicator of the presence of a pro-tumor immune response. Also provided are assay kits for determining an amount of mononuclear cell phenotype in performing the methods according to the present invention.

Provided is a magnetic separation device comprising a container having one or more outer surfaces; at least one flexible magnetic sheet; and a non-permanent adhesive that is used to detachably secure an outer surface of the container to a flexible magnetic sheet. A method of using the magnetic separation device according to the present invention comprises obtaining a fluid containing a mixed population of biological molecules, from which it is desired to separate at least one subpopulation of biological molecules; mixing the fluid with a magnetic separation reagent; contacting the mixture with the fluid holding chamber of the magnetic separation device; incubating the mixture for a sufficient time to allow for complexes to form between the subpopulation of biological molecules and the magnetic separation reagent; positioning the magnetic separation device in a position that magnetically attracts the complexes towards the flexible magnetic sheet, and thereby holds them in position in the container; and removing the remainder of the fluid from the magnetic separation device.

The present invention is directed to the measurement of cell-associated interleukin-2 receptor .alpha. (IL-2R.alpha.) expression in solid non-lymphoid tumors, and the use of such measurement In prognosing the metastatic potential of the tumor, diagnosing the metastatic localization of non-lymphoid tumor, and aiding the monitoring of efficacy of anticancer therapy against metastatic cells of non-lymphoid tumor. The present invention also relates to the use of T-cell receptor (tumor specific TCR.beta. idiotype) in monitoring the efficacy of anticancer therapy against non-lymphoid tumors.
