The present invention provides a method and a reagent for measuring periostin contained in a sample with improved accuracy, a method for improving accuracy in measurement of periostin, and a method of testing for pulmonary fibrosis or interstitial pneumonia with improved accuracy. The antibody of the present invention binds to at least one region selected from the group consisting of an EMI region, an R1 region, an R2 region, and an R3 region of periostin or a cleavage product thereof. The method and the reagent for measuring periostin and the method for improving accuracy in periostin measurement of the present invention is characterized by detecting at least one region selected from the group consisting of an EMI region, an R1 region, an R2 region, and an R3 region of periostin. The method of testing for pulmonary fibrosis or interstitial pneumonia of the present invention includes the steps of a) measuring the amount or concentration of periostin in a sample derived from a subject, which measuring includes detecting at least one region selected from the group consisting of an EMI region, an R1 region, an R2 region, and an R3 region of periostin and b) comparing the amount or concentration of the periostin in the sample derived from the subject with an amount or concentration of periostin in a sample derived from a living body not suffering from pulmonary fibrosis and interstitial pneumonia.

An objective of the present invention is to provide a method of testing for bronchial asthma or chronic obstructive pulmonary disease, a method of screening for candidate compounds for treating bronchial asthma or chronic obstructive pulmonary disease, and a pharmaceutical agent for treating bronchial asthma or chronic obstructive pulmonary disease. The present invention identified genes whose expression levels varied between respiratory epithelial cells that had been stimulated by IL-13 to induce the goblet cell differentiation, and unstimulated respiratory epithelial cells. The respiratory epithelial cells were cultured according to the air interface method. The genes were revealed to be useful as markers for testing for bronchial asthma or chronic obstructive pulmonary disease and screening for therapeutic agents for such diseases. Specifically, the present invention provides methods of testing for bronchial asthma or chronic obstructive pulmonary disease and methods of screening for compounds to treat the diseases based on the comparison of the expression levels of marker genes identified as described above.

SCCA1 and SCCA2 genes, whose expression were greatly changed in a plurality of cells by stimulating respiratory tract epithelial cells with IL-4 or IL-13, were obtained as allergy related genes. Furthermore, elevation of blood level of expression products of these genes accompanying bronchial asthmatic attack was elucidated. This invention provides a method of testing for bronchial asthmatic attack that uses as indicators, expression levels of these genes in biological samples.
