A dry multilayer analytical element for assaying transaminases wherein the analytical element comprises:a) a transaminase substrate;b) reagents that generate pyruvate in the presence of a transaminase and the transaminase substrate;c) pyruvate oxidase and cofactors for pyruvate oxidase; andd) reagents that generate a color in proportion to a concentration of hydrogen peroxide;and wherein the analytical element has, in the following order, a spreading layer, a reagent layer, and a support whereini) the spreading layer contains the transaminase substrate and a material selected from the group consisting of polymer beads, barium sulfate, titanium oxide, and mixtures thereof;ii) the reagent layer contains the pyruvate oxidase; andiii) both a hardened gelatin layer and a subbing layer separate the reagent layer from the spreading layer.

A multilayer analytical element has been prepared for accurate and rapid colorimetric determination of ethanol in aqueous specimens using alcohol dehydrogenase and an oxidized nicotinamide coenzyme. The element includes at least one reagent layer beneath a porous spreading layer. This reagent layer has a crosslinked polymeric binder prepared, in part, with a polymerizable monomer having a halomethylcarbonyl, haloethylcarbonyl, halomethylsulfonyl or haloethylsulfonyl group. Alcohol dehydrogenase is in a layer of the element for reaction with the analyte.

A multilayer analytical element has been prepared for accurate and rapid colorimetric determination of ethanol in aqueous specimens using alcohol dehydogenase and an oxidized nicotinamide coenzyme. The element includes three reagent layers beneath a porous spreading layer. The middle reagent layer has a crosslinked hydrophilic binder, while the other two reagent layers have uncrosslinked binders. In addition, both uncrosslinked reagent layers include a relatively high amount of a buffer having a primary amine, which buffer maintains the layer pH at from about 8 to about 10. Alcohol dehydrogenase is in the reagent layer next to the porous spreading layer.
Abstract:
Improved dry-operative ion-selective electrodes and their use are described. The dry-operative ion-selective electrodes include an internal reference electrode comprising a water-soluble salt dispersed in a polymer binder consisting essentially of a monomer having at least one carboxyl group and a hydrophobic monomer. The polymer provides reduced brittleness, good interlayer adhesion and high salt tolerance.
Abstract:
An analytical element for quantitatively assaying an analyte by measuring reflection density in the range 320 to 360 nm. The element comprises a reagent layer and a registration layer. One of such layers contains a dye which has a low absorbtivity band from 320 to 360 nm and a high absorbtivity band from 640 to 720 nm.
Abstract:
A colorimetric assay for the determination of conjugated, unconjugated or total bilirubin in biological fluids can be carried out with an improved analytical element. This element comprises a support having thereon a substantially gelatin-free mordant layer comprising a positively-charged interactive mordant having at least one binding site for bilirubin, a radiation-blocking layer, and a porous spreading layer. This element does not contain any colorimetric or fluorometric interactive compositions for bilirubin other than the mordant.

An overcoat composition for use in ion-selective electrodes comprises: (a) a discontinuous liquid phase comprising an oleophilic solvent dispersed within a continuous phase comprising a hydrophilic binder; (b) a complexing agent useful for extracting oleophilic anions; (c) a buffer which provides a pH in the range of from about 7.5 to about 9.5 under conditions of use; and (d) a nucleating agent. This overcoat composition is particularly useful in ion-selective electrodes for determination of CO.sub.2.
