摘要: An apparatus for culturing cells includes a bioreactor. The bioreactor may be modular and may include in a chamber a fixed bed, such as an unstructured or structured fixed bed (such as a spiral bed) for culturing cells, with a return column arranged centrally within the chamber. The modular bioreactor may include a plurality of structured fixed bed arranged in a stacked configuration. The modular bioreactor may include an outer casing forming a space for conditioning (e.g., insulating, heating, cooling) at least a chamber in which cells are cultured. The bioreactor may also include an impeller with radially curved blades, and may also suspend the impeller so that it may move from side-to-side and align with an external drive. Related methods are also disclosed.

The movement of the exit means from the closed position to the open position is controllable by said process control device.

摘要: A system for producing and purifying target biologics is disclosed. A cell culture unit is provided for generating target biologics in a volume of fluid. A purification unit is also provided for purifying the target biologics in the volume of fluid portion directly from the cell culture unit without a clarifying step, such as by using one or more separators, such as expanded beds or chromatography columns arranged in parallel, and potentially operating in series to achieve continuous operation. The one or more separators may include magnetic beads adapted for binding to the target biologics. The purification unit may receive at least a portion of the volume of fluid directly from the cell culture unit, and a collection unit may be provided for collecting the purified target biologics from the purification unit, and also may be adapted for returning the volume of fluid without the target biologics to the cell culture unit. The system may comprise a first vessel adapted to concentrate a volume of fluid including the target biologics, and a second vessel adapted to further concentrating the volume of fluid, the first or second vessels including magnetic beads adapted for binding with the target biologics. Related methods are also disclosed.

摘要: An apparatus for sampling a cell culture includes a sampler for connecting to a bioreactor with a fixed bed including the cell culture. A releasably attached sample portion for positioning within or adjacent the fixed bed includes a substrate on which cells may be grown and then removed from the bioreactor to provide an indication of cell growth. A kit for sampling a cell culture bed of a bioreactor includes the sampler for sampling the cell culture bed with the releasably attached sample portion, along with a container adapted to detach the sample portion from the sampler. A method of obtaining a sample of a cell culture in a bed within a bioreactor includes positioning a sampler positioned at least partially in or adjacent to the bed to promote the growth of cells on a releasably attached portion of the sampler, withdrawing the sampler from the bioreactor, and detaching the portion of the sampler.

摘要: The current disclosure concerns a method for decontaminating a virus production system, wherein virus particles are produced in at least one bioreactor and purified from liquids comprising said virus particles thereby generating liquid waste, said method comprising the step of exposing said system or a part thereof to heated air and/or liquid at a temperature of at least 60° C. after viral production took place, thereby eradicating remaining virus particles in said system. In a second and third aspect the current disclosure concerns a virus production system wherein the system comprises a heating device suitable for heating air and/or liquid to a temperature of at least 60° C., and the use of such a system for the production of viruses and/or viral vaccines.
