    摘要: In a database MSn spectrum search, search of a known compound whose principal chain is identical to that of an unknown compound is enabled, thereby allowing analysis of an entire structure even if the entire structures of the known compound in database and that of the measured unknown compound are not identical. The MSn spectrum obtained in the MSn measurement of the unknown compound is compared with all the MSm spectra (m≧1) in the database regardless of MSn generation. In the MSn measurement of a series of related compounds including various types of different side chains of the same principal chain, such as in the case of biopolymers, it becomes possible to determine the structure of the principal chain using a database search even if the entire structure is not clear. And the estimation of the entire structure is made possible on the basis of the principal chain structure.

In a mass spectrometer with an MSn analysis function for ionizing a sample eluted from a separation unit for separating the sample into individual components, fragmenting an ion of a desired mass number, and performing a mass analysis of fragment ions, the mass spectrometer includes a database storing correlation information between an isomer presence ratio and a particular ion intensity ratio in a mass spectrum per isomer. An isomer ratio can be clarified even for a mixed sample of isomers, such as enantiomers, which are difficult to separate from each other by the separation unit and cannot be discriminated by MS.

    摘要: Provided are a novel marker of the state of cells and the differentiation level of the same which is usable as a substitute for a gene or a protein; a means for quantitatively understanding this marker; and a means for understanding the state of specific cells and the differentiation level of the same using this marker. A method for determining a sugar chain category to be used for evaluating the state of cells. The quantitative profile of N-linked sugar chains of cells before a change in the state thereof is obtained. Next, the quantitative profile of N-linked sugar chains contained in the cells after the change in the state thereof is obtained. Sugar chains showing variations in the contents thereof between these two quantitative profiles are extracted and the individual sugar chains contained in the extracted sugar chains are classified into the following categories based on sugar chain type: either the high-mannose type or the non-high-mannose type; (1) the presence/absence of a bisect; (2) the number fucose residues (0, 1, or 2 or more); (3) the presence/absence of sialic acid; and (4) the degree of branching (2 or 3 or more). From these five categories, a sugar chain category appropriate for evaluating the state of cells is determined. A method for evaluating the state of cells and the differentiation of cells using the thus determined sugar chain category.

    摘要: A method of analyzing a sugar chain by high performance liquid chromatography uses a column packed with a stationary phase including anion and cation exchangers. A fluorescence-labeled sample is prepared for the analysis by decomposing glycosaminoglycans to be analyzed into their constitutional units being disaccharides, capturing the disaccharides by glycoblotting, releasing the disaccharides by acid treatment, and subsequently reductively aminating the released disaccharides. In this method, two or more types of glycosaminoglycans can be simultaneously analyzed.
