A reflector is provided as well as a time-of-flight mass spectrometer with a reflector. The mass spectrometers is used for determining the chemical structure of molecules as well as for the quantitative analysis of unknown mixtures of substances. Design effort is minimized, especially for the reflector. The reflector is present in the time-of-flight mass spectrometer to generate an electrostatic field permitting the best possible focusing for the deflection of the ions. The reflector body is made in one piece as a radially symmetrical trough. The reflector is preferably made of a stainless steel or a carrier material with conductive coating and is polished on the inner side of the trough.

A hybrid mass spectrometer comprises an ion mobility (IMS) section and a mass analysis section that analyses ions based on mass-to-charge ratios. In the IMS section, a DC potential gradient is established and a drift gas provided, so as to separate ions based on varying ion mobilities. Additionally, at least a downstream portion of the IMS section includes a rod set focusing ions along the axis, this prevents loss of ions and gives good transfer of ions into-a-mass analysis section, which can be a time-of-flight mass analyzer or an analyzer including a quadrupole rod set. A collision cell and mass analyzer can be provided between the two sections for MS/MS analysis. The IMS section then provides better utilization of an available sample; as each group of ions is elected from the IMS section, one ion can be mass selected as a precursor, for subsequent fragmentation/reaction and subsequent mass analysis of the product ions. Another aspect of the invention provides the ability to form potential wells, utilizing a segmented rod set configuration, so as to trap and hold ions after separation based on mobility characteristics.

The present invention relates to a method of separating ions having the same mass to charge ratio but different ion mobility characteristics, based on the ion focussing principles of high field asymmetric waveform ion mobility spectrometry. Isomers including gas-phase molecular anions of the amino acids leucine and isoleucine are separable by the method of the present invention. Identification of different conformers of ions at the same charge state, including conformers present at certain charge states of the protein bovine ubiquitin, is also possible using the method of the present invention.

One illustrative embodiment of a voltage pulser circuit comprises a voltage source producing a first voltage, and a thyratron tube having an anode coupled to the output of the voltage source, a cathode connected to a reference potential and a grid responsive to a grid control voltage to electrically connect the anode to the cathode to thereby cause the first thyratron tube to switch the anode between the first voltage and the reference potential. A pulse-shaping circuit may be connected to the anode of the tube to effectuate desired rise and fall times of the voltage pulses produced by the voltage pulser circuit. Such a voltage pulser circuit is particularly suited for use in connection with the operation of pulsed spectrometer instruments, such as time-of-flight mass spectrometers and the like.

A Time-of-Flight Mass Spectrometer (TOF-MS) is configured to improve resolution and sensitivity performance. The TOF-MS includes an arrangement of electrodes comprising an ion accelerator with two stages of homogeneous electric fields, an ion reflector with a single stage of a homogeneous electric field, accelerator and reflector being separated by a first drift space, and an ion detector which is separated from the reflector by a second drift space. Contrary to known TOF-MS of similar configuration, the set of electric potentials which must be applied to said electrodes is predetermined for a given geometry in such a way that a spatial distribution of ions initially at rest in the first gap of the said accelerator is compressed at the location of the detector in the longitudinal direction to a focus of first and second order in the initial axial coordinate. Therefore, mass resolution is enhanced over a TOF-MS that provides only for longitudinal focusing of first order, while the number of passages through grid electrodes along the flight path is reduced, and hence ion transmission and instrument sensitivity are improved.

A time-of-flight mass spectra calibration technique uses time-of-flight mass spectrometer instrument operational parameters and known mass and measured time-of-flight data pairs to optimize values of chosen ones of the instrument operational parameters. Electrostatic time-of-flight calculations are conducted in conjunction with an iterative procedure, preferably a simplex optimization procedure, to thereby minimize a residual error between the electrostatic time-of-flight calculations and the measured time-of-flight data values for each of the known mass values. While conventional curve fitting mass calibration techniques are devoid of information that describe ion behavior, the mass calibration technique of the present invention, by contrast, takes into account all of the instrument operational parameters in arriving at a final calibration. Because the electrostatic TOF calculation is a description of ion behavior in an actual TOF mass spectrometer instrument rather than a polynomial representation of a curve, it is well behaved and does not contain any instabilities where unpredictable calibration errors might occur. Moreover, unlike conventional curve fitting mass calibration techniques, the mass calibration technique of the present invention maintains mass accuracy in extrapolated mass ranges.

Various configurations of characterization systems such as ion mobility spectrometers and mass spectrometers are disclosed that are coupled to an ionization device. The ionization device is formed of a membrane that houses electrodes therein that are located closer to one another than the mean free path of the gas being ionized. Small voltages across the electrodes generate large electric fields which act to ionize substantially all molecules passing therethrough without fracture. Methods to manufacture the mass spectrometer and ion mobility spectrometer systems are also described.

A mass spectrometer is disclosed wherein ions from a pulsed ion source are dispersed in a drift region so that the ions become separated according to their mass to charge ratios. The ions are then received by an ion guide in which multiple trapping regions are created and wherein the multiple trapping regions are translated along the length of the ion guide. The ion guide receives the ions so that all the ions trapped in a particular trapping region have substantially the same or similar mass to charge ratios. The ions are released from the exit of the ion guide and the pusher/puller electrode of an orthogonal acceleration Time of Flight mass analyzer is arranged to be energised in synchronization with the ions emerging from the ion guide. The trapping regions may be translated along the ion guide with a velocity which becomes progressively slower and the delay time of the pusher/puller electrode may be progressively increased.
