The present invention relates to the generation of chimeric antigen receptors (CAR) referred to as multi-chain CARs. Such CARs, which aim to redirect immune cell specificity and reactivity toward a selected target exploiting the ligand-binding domain properties, comprise separate extracellular ligand binding and signaling domains in different transmembrane polypeptides. The signaling domains are designed to assemble in juxtamembrane position, which forms flexible architecture closer to natural receptors, that confers optimal signal transduction. The invention encompasses the polynucleotides, vectors encoding said multi- chain CAR and the isolated cells expressing them at their surface, in particularly for their use in immunotherapy. The invention opens the way to efficient adoptive immunotherapy strategies for treating cancer and viral infections.

The present invention relates to a medium composition for culturing self-activated lymphocytes applicable to the treatment of a malignant tumor, and a method for culturing self-activated lymphocytes using the same, and more specifically, to a medium composition for culturing self-activated lymphocytes which effectively amplifies and activates natural killer (NK) cells, T cells and NKT cells by adding anti-CD3 and anti-CD16 antibodies in addition to interleukin-2 (IL-2), interleukin-12 (IL-12) and interleukin-18 (IL-18) to a medium, and can culture immunocytes which are remarkably effective for the treatment of various types of malignant tumors by increasing the relative ratio of NK cells among lymphocytes by leaps and bounds, and a method for culturing self-activated lymphocytes using the same. According to the present invention, the method for culturing self-activated lymphocytes comprises the following steps: extracting lymphocytes from peripheral human blood; first, culturing the extracted lymphocytes in a culture solution containing interleukin-2 (IL-2), L-glutamine and self-induced plasma in the presence of interleukin-12 (IL-12), interleukin-18 (IL-18), and anti-CD3 and anti-CD16 antibodies; and mixing the culture mixture solution cultured in the first culturing step with a culture solution containing IL-2, L-glutamine and self-induced plasma and second, culturing the obtained mixture solution in the presence of IL-12, an anti-CD16 antibody and IL-18.

Also provided are methods for isolating the subpopulation of bone marrow-derived adherent stem cells from bone marrow-derived adherent cells and for using the isolated subpopulation of bone marrow-derived adherent stem cells for treating tissue and/or organ damage in a subject.
