The invention provides polynucleotide constructs for the regulation of gene expression by aptamer-based modulation of self-cleaving ribozymes and methods of using the constructs to regulate gene expression in response to the presence or absence of a ligand that binds the aptamer. The invention further provides methods for making and using riboswitches that decrease target gene expression in response to an aptamer ligand as well as riboswitches that increase target gene expression in response to an aptamer ligand.

Specifically, the present invention provides a polymer comprising rigorously alternating AB subunits and methods of formation of the AB alternating polymers. In the polymers and process of the invention, the A monomer is derived from a cyclobutene derivative, and the B monomer is derived from a cyclohexene derivative. The polymerization takes place in the presence of an olefin metathesis catalyst.

A method and apparatus for determining a temperature differential at a portion of a patient's body utilizing temperature sensitive MRI measurements. A diagnostic fluid bolus is administered into a blood vessel of the patient, wherein the diagnostic fluid bolus has a diagnostic fluid bolus temperature waveform. MRI measurements are used to determine the thermodiluted temperature waveform of the diagnostic fluid bolus at a target site in the body spaced away from the administration site. The temperature differential may be used to determine a cardiovascular parameter.
