Synthesis of semi-synthetic monobactamic or beta -lactamic antibiotics by using derivatives stabilized by various methods of penicillin G acylase from various microbial sources according to a thermodynamically controlled strategy in monophase water/cosolvent organic apolar systems, wherein the concentration of the cosolvent varies between 30 % and 90 %, the temperature between -10 DEG C and 50 DEG C, the pH between 4.5 and 8.5, with concentrations of the antibiotic nucleus between 0.5 and 875 mM and acyl donor between 0.2 mM and 1 M, with a relationship antibiotic ring/activated or free acyl donor, using a buffer between 0 and 1 M. A process for the preparation of cephalosporin acid derivatives comprises the step of enzymatically converting cephalosporin C into 7 cephalosporin acid derivatives in the presence of two biocatalysts, wherein one biocatalyst comprises co-immobilised D-amino acid oxidase and a catalase enzyme, and the other biocatalyst comprises immobilised glutaryl acylase.

Abstract:
The method is essentially based on the reaction of ethylenediamine with inert supports, containing a controlled single layer of similar aldehyde groups which are moderately separated from the support surface and on the following reduction by using borohydride of the activated supports in order to eliminate the reactive additional groups which could otherwise cause secondary indesired interactions. Applications to biotechnology, specially for the purification of proteins and for the preparation of fixed protein derivatives.

El proceso de inmovilización de enzimas y proteínas sobre este tipo de soporte tiene lugar en dos etapas consecutivas: a. una primera adsorción iónica muy rápida de la enzima sobre el soporte ionizado en condiciones experimentales muy suaves y b. la inmovilización covalente multipuntual intensa entre la enzima previamente adsorbida y una muy alta densidad de grupos epóxido presentes en el soporte. Las principales características prácticas de este nuevo método de inmovilización son: la inmovilización muy rápida de grandes cantidades de enzima en condiciones experimentales muy suaves, la posibilidad de conseguir la estabilización de las enzimas inmovilizadas por inmovilización covalente multipuntual al soporte, la no utilización de ningún reactivo inestable ni tóxico durante o después de la inmovilización, etc.
