The present invention provides compositions and methods for the production of glycoproteins with enhanced sialylation. In particular, the invention provides cell lines comprising disrupted sialidase expression and methods of using the cell lines to produce glycoproteins with enhanced sialylation.

Disclosed is an air volume calculation system for a variable speed or constant speed fan, comprising a central processing control system, an upper processing system, a moving vane angle indication transmitter (3) of the fan, an entrance pressure transmitter (14) of the fan, an entrance air temperature transmitter (15) of the fan, an exit pressure transmitter (16) of the fan, a motor current transformer (17) of the fan and a DCS control system (11) of the fan; wherein the moving vane angle indication transmitter (3) of the fan, the entrance pressure transmitter (14) of the fan, the entrance air temperature transmitter (15) of the fan, the exit pressure transmitter (16) of the fan and the motor current transformer (17) of the fan are separately connected to the signal input end of the central processing control system; and the signal input end of the central processing control system is connected to the DCS control system (11) of the fan. The system can decrease the installation complexity of the air volume measurement device, reduce the resistance loss caused by the air volume measurement device, reduce the air volume measurement error, increase the reliability of safe running and add an online air volume characteristic monitoring function.

A strain of xylose-utilizing Zymomonas was engineered with a genetic modification to the glucose-fructose oxidoreductase gene resulting in reduced expression of GFOR enzyme activity. The engineered strain exhibits reduced production of xylitol, a detrimental by-product of xylose metabolism.lt also consumes more xylose and produces more ethanol during mixed sugar fermentation under process-relevant conditions.

A micro flow cytometer having the ability to distinguish light emanating from a particle in the interrogation zone (6) of the cytometer contains a plurality of optical waveguides (10, 11, 12, 13) for receiving emanating light, the optical waveguides (10, 11, 12, 13) positioned at different angles from the axis of the interrogation zone and in the same plane. Photodetectors measuring emanated light provide an electrical signal which is processable to reveal an angular dependence of wavelength with respect to the interrogation channel (6).

Identified are mutants of the promoter of the Z. mobilis glyceraldehyde-3-phosphate dehydrogenase gene, which direct improved expression levels of operably linked heterologous nucleic acids. These are high expression promoters useful for expression of chimeric genes in Zymomonas, Zymobacter, and other related bacteria.

Strains of Zymomonas were engineered by introducing a chimeric xylose isomerase gene that contains a mutant promoter of the Z. mobilis glyceraldehyde-3-phosphate dehydrogenase gene. The promoter directs increased expression of xylose isomerase, and when the strain is in addition engineered for expression of xylulokinase, transaldolase and transketolase, improved utilization of xylose is obtained.

Through screening of a Zymomonas mutant library the himA gene was found to be involved in the inhibitory effect of acetate on Zymomonas performance. Xylose-utilizing Zymomonas strains further engineered to reduce activity of thehimA gene were found to have increased ethanol production in comparison to a parental strain, when cultured in mixed-sugars medium comprising xylose, and, in particular, in the presence of acetate.
