摘要: The invention relates to self-assembling peptide (nano)fibers, hydrogels, and methods, uses and intermediate products and kits relating thereto. Provided is a method for providing peptide-based functionally modified (nano)fibers, comprising (i) providing a fiber forming solution comprising pseudopeptide building blocks of the formula A-Peptide-B, wherein: Peptide is a moiety of 1 to 8 amino acid residues having a sequence that is predisposed to form a one-dimensional array, such as β-sheet fibrils; A is an aromatic moiety carrying two reactive thiol groups; and B is a reactive α-nucleophile; (ii) exposing the fiber forming solution to oxidizing conditions to induce supramolecular self-assembly of the pseudopeptide building blocks into peptide-based (nano)fibers; and (iii) contacting said (nano)fibers with at least one biologically relevant functional group of interest comprising reactivity C forming a reactive pair with B to obtain covalently functionally modified nanofibers.

摘要: Vaccines directed to the SARS-CoV-2 antigen, methods of administering and/or treating a subject (including prophylactic treatment) infected and/or at risk of being infected with SARS-CoV-2. A transdermal membrane comprising a non-infectious icosahedral phage vaccine displaying a SARS-CoV-2 antigen is described wherein the membrane is stable at room temperature for greater than 3 months and uses thereof to vaccinate a subject against the SARS-CoV-2 antigen.

摘要: Proteins and fusion proteins for forming menbraneless droplets in cells are provided. Described herein, is the development of a protein, named PopTag, that drives phase separation when it is part of a chimeric fusion protein. PopTag is engineered from the PopZ protein, found in a-proteobacteria (including Caulobacter crescentus). Despite PopZ being exclusively found in this clade of bacteria, the PopTag can drive protein phase separation in other prokaryotes (e.g., E. coli) and eukaryotes (e.g., human cells).

摘要: The present invention provides a synthetic, modular system for DNA modification as may be employed for genome editing comprising a targeting nucleic acid possessing both DNA targeting ability and ability to bind a recognition module of a modular polypeptide component, where the modular polypeptide component also includes an effector component and a short peptide DNA-binding sequence (DBD) which binds a pre-determined sequence (PBS) in the target. The DBD may preferably be no more than a 15 mer and serves to destabilize the structure of a targeted dsDNA upon binding thereby facilitating the desired DNA modification. The effector component may, for example, be an artificial nickase comprising linked self-assembling peptides thereby providing a complete genome-editing system of advantageous small size for vector delivery to cells.

摘要: Disclosed are methods of a method of making a nanostructure, comprising adding a component A (compA) protein to a solution comprising a component B (compB) protein under conditions that minimize shear stress, thereby forming a compA:compB complex. Further disclosed are methods of making a nanostructure, comprising (i) providing a first inlet fluid stream comprising a first protein and a second inlet fluid stream comprising a second protein, and (ii) contacting the first inlet fluid stream and the second inlet fluid stream to form an outlet stream, wherein mixing of the first protein and the second protein occurs in the outlet stream, thereby forming a protein complex comprises the first protein and the second protein. A microfluidic mixer may be used. The methods may further comprise purifying the compA:compB complex from excess compA, excess compB, and/or other impurities by filtering the solution with a 1,000 kDa membrane or an equivalent thereof.
