摘要: This technology provides a liquid dispensing system with a liquid loading device that can be used to load small volumes of liquid by sliding over or under a surface-patterned substrate, or, alternately, moving the surface-patterned substrate over or under the liquid loading device. The surface-patterned substrate may be a substrate with patterned geometry or wettability. The liquid loading device spatially confines liquid against the surface-patterned substrate and delivers it to intended areas on the surface-patterned substrate. The liquid loading device accomplishes this by maintaining liquid in spatially confined sections, and moving relative to the substrate, by, for example, sliding over it to deposit liquid on the surface-patterned substrate. The liquid loading device utilizes geometry and/or wettability to achieve liquid confinement and delivery to intended areas on the surface-patterned substrate.

摘要: Methods for identifying a cell population secreting a biomolecule with desirable attributes are disclosed. The desirable attributes can include, for example, quantity and quality (e.g., minimal aggregation and/or desired configuration). Cell populations identified by the disclosed methods are more likely to successfully scale during production. The methods can include assessing multiple domains/binding sites of a complex biomolecule and/or the formation of aggregates by the biomolecules. Methods of assessing a secretion level for biomolecules having a wide range of molecular weights and method for enhancing loading of cells into chambers of a microfluidic device. The labelling of viable cells and/or cells actively expressing a biomolecule of interest, for example, can permit selection and subsequent analysis of the cells most likely to successfully expand and express the biomolecule of interest, thereby reducing effort, risk, and cost associated with the screening of cells and increase the probability of identifying cell lines that are optimal producers of biomolecules of interest.

摘要: The present invention provides methods and systems for analyzing a biological sample. The methods of the present invention can utilize a labeling moiety and a cleavage moiety. The labeling moiety can comprise a binding moiety that is coupled to a detectable tag via a polynucleotide linker. The cleavage moiety can form a complex with the polynucleotide linker. In some cases, after formation of the complex, the cleavage moiety can effect cleavage of the polynucleotide linker, to release the detectable tag from the binding moiety. The release of the detectable tag can allow one or more additional imaging of the biological sample with a different labeling moiety that comprises a different binding moiety but the same labeling moiety.

摘要: Disclosed herein are compositions and methods for attenuating stress-induced mental fatigue in subject in need thereof by administering to the subject a composition comprising from about 2 mg to about 800 mg of paraxanthine. In certain embodiments, paraxanthine is present in the composition in amount from about 20 mg to about 600 mg. In further embodiments, paraxanthine is present in the composition in amount from about 50 mg to about 400 mg. According to certain embodiments, fatigue attenuated by the instantly disclosed method is a result of physical stress on the subject. Physical stresses that may result in mental fatigue include, but are not limited to, periods of intense exercise.
