A novel cytokine designated TRAIL induces apoptosis of certain target cells, including cancer cells and virally infected cells. Isolated DNA sequences encoding TRAIL are disclosed, along with expression vectors and transformed host cells useful in producing TRAIL polypeptides. Antibodies that specifically bind TRAIL are provided as well.

Methods of production of desired recombinant proteins, polypeptides and peptides are disclosed which utilise mammalian host cells engineered for autonomous and regulated growth in low cost protein/serum-free culture media. Preferred host cells express insulin or an insulin-like growth factor, and/or transferrin and are engineered so that addition of an inducer (e.g. ZnCl2 + CdCl2) halts growth and simultaneously induces the expression of the desired recombinant protein, polypeptide or peptide.

The invention provides methods and compositions for targeting endocrine cells capable of converting an immature expressed protein into a mature protein, and secreting the expressed protein into the systemic circulation. In particular, the invention features methods and compositions for treating disorders, such as diabetes by orally administering a vector expressing insulin to target cells which can subsequently secrete the expressed insulin into the blood circulation.
