Various methods for evaluating hemostatic effect and various blood coagulation initiation reagents were studied to construct a method for evaluating blood coagulation reaction mediated by a substance having an activity of substituting for coagulation factor VIII (FVIII). As a result, it was discovered that by using a coagulation initiation reagent containing activated coagulation factor XI (FXIa) and phospholipids, the effect of a substance having an activity of substituting for coagulation factor VIII (FVIII) on blood coagulation reaction can be evaluated using the amount of thrombin generated in the blood sample as an indicator.

The methods comprise examining circulating endothelial cells from the subject for the presence of one or more coagulation factors and correlating an amount of circulating endothelial cells exhibiting one or more coagulation factors with the level of endothelial disease in the subject. Anti-coagulation factor therapy can then be administered to the subject to address the endothelial disease.

In some embodiments, the invention provides methods for detecting an aberrant fibrinolysis condition in a patient. The method includes subjecting a blood sample from the patient to a viscoelastic analysis in the presence of a known amount of a thrombolytic agent, to obtain a coagulation characteristic value of the patient; and comparing the coagulation characteristic value of the patient to a coagulation characteristic value of a healthy individual, the coagulation characteristic value of the healthy individual obtained by subjecting a blood sample from a healthy individual to the viscoelastic analysis in the presence of the known amount of the thrombolytic agent, wherein a difference in the coagulation characteristic value of the patient as compared to the coagulation characteristic value of the healthy individual identifies the patent as having aberrant fibrinolysis. In some embodiments, the invention also provides a container adapted for detecting an aberrant fibrinolysis condition in a blood sample using viscoelastic analysis comprising an interior having a coating comprising a thrombolytic agent.

The present invention provides a method of detecting platelet activation in a patient, the method comprising the steps of a) obtaining a blood sample from a patient suspected of having heparin-induced thrombocytopenia (HIT); b) incubating an effective amount of platelet factor 4 (PF4) with a sample of platelets to yield a sample of PF4-treated platelets; c) contacting the patient blood sample with the PF4-treated platelets; and d) measuring the extent of platelet activation, wherein an increase in platelet activation compared with results obtained using a normal blood sample is indicative of the patient having HIT.

The fluidics device further includes at least one fluid channel configured to accept the array. The fluidics device can further include a measuring element configured to measure a degree of deflection of one or more of the flexible posts in the array. In some embodiments, the fluidics device comprises a handheld device and usable for point of care testing of platelet forces and coagulation.

The present disclosure provides methods and compositions for diagnosing and treating subject having a bleeding disorder. The disclosed methods comprise contacting a sample, e.g., a blood or plasma sample obtained from the patient, with an activation mixture comprising an activated coagulation factor and a phospholipid mixture, wherein the activation mixture is dried onto a solid substrate. Also provided is a global hemostasis test based on the integration of clotting time (Ct) and pharmacokinetics data. The methods and compositions presented can be applied to point-of-care diagnostic systems.
