    摘要: Disclosed are fusion proteins comprising two detection portions and a periplasmic binding protein (PBP) portion between this two detection portions which undergoes a conformational change upon binding of a compound. Also described are nucleic acid molecules comprising a nucleotide sequence encoding such a fusion protein as well as expression cassettes, vectors, transgenic plants and transgenic non-human animals comprising said nucleic acid molecules. In addition, described are methods for detecting an analyte in a sample or in a cell using said fusion protein, and optionally, a control sensor allowing a calibration of analyte detection made by said fusion protein. Also described are such control sensors and nucleic acid molecules encoding them, as well as diagnostic compositions, kits and uses of the fusion protein and the control sensor for in vitro or in vivo analyte detection and preparing a diagnostic composition.

    摘要: A process for inducing a transcriptionally active chromosome (i.e., a 'lampbrush' chromosome) from condensed chromatin or nucleus is disclosed. The condensed chromosome is contacted with the contents of a germinal vesicle. Preferably, the nuclear envelope is disrupted or removed. Moreover, a heterologous system of chromosome and germinal vesicle derived from organisms of different species is preferred because it permits analysis of organisms that do not have lampbrush chromosomes or cannot be manipulated by other techniques. Such lampbrush chromosomes can be attached to a substrate, and then analyzed by a variety of molecular and cytological techniques such as, for example, antibody detection of chromosomal protein, autoradiography, electron and light microscopy, histochemistry, image analysis, immunofluorescence, in situ hybridization of nucleic acids, morphology, and the like.

    摘要: An apparatus (10) is provided for detecting deformation in solids in deep holes having a resilient vessel (12) pre-stressed into a rock formation. The vessel (12) is filled with fluid (14) and is sealed except for an outlet tube (16) of small diameter which extends all the way to, or close to, the surface (18). There the outlet tube (16) terminates in an extensible bellows (20). The whole apparatus is completely filled with fluid (14). Any deformation of the vessel (12) will cause fluid to be forced through the outlet tube (16) and so cause the bellows (20) to extend or contract. This motion can be monitored by any position-sensitive transducer (22) such as a linear variable differential transformer or capacitance bridge. A second tube (24) of the same length and size as the outlet tube (16) is filled with the same fluid. This tube (24) terminates at the bottom in close proximity to the resilient vessel (12). It also has a bellows (26) and a displacement transducer (28) at its upper end at the surface (18).
