    摘要: A method and reagent system for the rapid isolation, identification, and/or analysis of leukocytes from a whole blood sample. The reagent system includes a lytic reagent solution comprising an aqueous solution of a water soluble compound which at least partially dissociates in aqueous solution to release a proton and a counterion and which, upon contact with a blood sample, acidifies the sample to a pH in the range of from about 2.6 to 4.0. The reagent system also includes a quenching reagent solution comprising an aqueous alkaline salt solution which effectively and rapidly arrests the chemical action of the lytic reagent solution on the sample and restores the native physiological environment of the sample. The method comprises using the reagent system to selectively lyse the red blood cells in a blood sample and modify the leukocytes to enable their differentiation into five sub-populations.

    摘要: A particle analyzing method and apparatus for white blood cell (leukocyte) differentiation by optical and electronic measurements, without the use of dyes, on a flowing stream of leukocytes. A hydrodynamically focused stream of the leukocytes, in native condition, is passed into and through a beam (20) of radiant energy in a flow cell (10), whereby the beam is uniquely scattered by different ones of the subpopulations of the leukocytes. The scattered light impinges upon light collection means (24) disposed adjacent the flow cell. For eosinophil detection, the scattered light is collected at angular positions from about 10° to 70° relative to the axis (26) of the beam (20). Other of the leukocyte subpopulations are differentiated relative to the eosinophils and each other by optical and/or electronic measurements, especially including the Coulter Principle of particle analysis.

    摘要: This invention primarily is directed to a hematology reference control solution, the three separate white cell controll portions thereof consisting of three types of fixed red cells of determined size distribution for checking the operation of a particle analyzing instrument, including its predetermined lower and upper threshold settings for each class or subclass of leukocytes. For preparing a human granulocyte analogue, nurse shark erythrocytes are altered and fixed in a chilled solution to simulate in number, size and distribution the granulocytes in human whole blood.     摘要: An improved procedure for statistically evaluating the performance, precision and/or accuracy, of at least one analytical instrument in a pool of like or substantially like instruments by comparing the performance of the one instrument located in one laboratory with the performance of like instruments in other laboratories. The procedure includes the step of quantitatively analyzing a reference or control specimen of known constituents by each instrument in the pool and collecting the resultant data from the one instrument in the pool and of the pool instruments. The mean data from the one instrument and of the pool instruments are mathematically correlated in a series of statistical equations for computing a novel, sensitive index known as the "Instrument Performance Index" or IPI. This IPI can be tabulated and/or graphically displayed, preferably as a single point in a Cartesian coordinate system, whereby an operator can tell at a glance the performance level with respect to both precision and accuracy of the one instrument in the pool. The ultimate IPI is composed of a novel Coefficient of Variation Index (CVI), which is a valuable measure of instrument precision, and the Standard Deviation Index (SDI), which is a measure of performance accuracy. Both the CVI and SDI are derived values according to the invention and are capable of tabulation and display.
