摘要:
Nanoprisms containing silver and gold are disclosed. The nanoprisms exhibit the properties of pure silver nanoprisms, but are less susceptible to silver modification or reaction by a surrounding environment than pure silver nanoprisms due to the presence of the gold. The gold surface of the nanoprisms can be further modified, using known gold-modification techniques.

The present invention relates to compositions and methods for altering (e.g., enhancing) RNAi. In particular, the present invention provides systems and methods for identifying regulators of RNAi. For example, the present invention provides RNAi regulators (e.g., HPSl and HPS4) and methods of altering (e.g., inhibiting) these regulators in order to alter (e.g., enhance) RNAi. The present invention also provides methods of identifying inhibitors (e.g., small molecule, nucleic acid (e.g., siRNA), and antibody) of RNAi regulators and methods of using the same (e.g., to enhance RNAi). Compositions and methods of the present invention find use in research (e.g., functional genomics), therapeutic (e.g., drug discovery and delivery) and clinical applications.

The present invention relates to the use of stem cell factor (SCF) polypeptide, alone and in combination with granulocyte colony stimulating factor (GCSF) polypeptide, in the prevention or treatment of injury to the brain after cerebral ischemia or neurological disorder. More particularly, the invention provides methods of improving neurological function and outcome after stroke by the administration of SCF polypeptide, alone and in combination with G-CSF polypeptide. This treatment can be used alone or in combination with other well-known methods of treatment of cerebral ischemia and neurological disorders in a mammal.

The invention provides methods of generating angiostatin in vitro comprising contacting plasminogen with a plasminogen activator and a sulfhydryl donor or contacting plasmin with a sulfhydryl donor. The invention also provides a method of treating angiogenic diseases by administering to an animal suffering from such a disease a sulfhydryl donor and, optionally, a plasminogen activator, plasminogen or plasmin. The invention further comprises a composition for generating angiostatin comprising a sulfhydryl donor and a plasminogen activator. The invention also provides a container holding a sulfhydryl donor and/or a plasminogen activator, said container having a label thereon instructing administration of the sulfhydryl donor and/or plasminogen activator to an animal suffering from an angiogenic disease. The invention further provides plasminogen fragments-whose N-terminal amino acid is the same as that of plasmin and whose C-terminal amino acid is located in kringle 5 and which inhibit angiogenesis, antibodies which bind selectively to these fragments, methods and kits for using the antibodies, methods and materials for making the fragments by recombinant DNA techniques, and a method of treating an angiogenic disease comprising administering an effective amount of one of the fragments. Finally, the invention provides a method of treating an angiogenic disease comprising administering a transgene coding for one of the fragments.

The invention provides amyloid beta-derived dementing ligands (ADDLs) that comprise amyloid β protein assembly bled into globular non-fibrillar oligomeric structures capable of activating specific cellular processes. The invention provides methods for assaying the formation, presence, receptor protein binding and cellular activity of ADDLs, as well as compounds that block the formation or activity of ADDLs, and methods of identifying such compounds. The invention further provides methods of using ADDLs, and modulating ADDL formation and/or activity, inter alia in the treatment of learning and/or memory disorders.

A method of nanolithography comprising: transporting a patterning compound from a nanoscopic tip to a substrate to form a pattern on the substrate, wherein the patterning compound has a first electrostatic charge and the substrate has a second electrostatic charge which is opposite to the first electrostatic charge. The patterning compound can be an electrically conductive polymer having a charged polymer backbone. The patterns can be dots and lines having lateral dimensions of less than one micron. No electrical bias from an external voltage source between the tip and the substrate is needed.

The use of direct-write nanolithography to generate anchored, nanoscale patterns of nucleic acid on different substrates is described, including electrically conductive and insulating substrates. Modification of nucleic acid, including oligonucleotides, with reactive groups such as thiol groups provides for patterning with use of appropriate scanning probe microscopic tips under appropriate conditions. The reactive groups provide for chemisorption or covalent bonding to the substrate surface. The resulting nucleic acid features, which exhibit good stability, can be hybridized with complementary nucleic acids and probed accordingly with use of, for example, nanoparticles functionalized with nucleic acids. Patterning can be controlled by selection of tip treatment, relative humidity, and nucleic acid structure.

Self-assembling compounds capable of forming birefringent gels when added to an organic medium in an amount of about 0.05 % to about 2 % by weight are disclosed. A birefringent gel containing a self-assembling compound and an organic monomer can be polymerized to produce an optically birefringent polymer. The birefringent polymer can be melted to provide a birefringent melt that then can be formed into an article of manufacture, like an optical fiber.
