Described are a therapeutic monoclonal antibody specific for the Type Six Secretion System (T6SS) needle protein of Acinetobacter baumannii (A. baumannii) and methods of use. Specifically, the antibody specifically binds to hemolysin co-regulated protein (Hep). Further disclosed are methods of using an anti-Hep antibody for detecting A. baumannii in a sample as well as a mutant A. baumannii strain comprising a deletion or mutation in its genome such that said deletion or mutation interrupts the proper translation of its hep protein.

The present invention is directed to recombinant DNA technology and immunology, particularly antigens and related vaccines for prevention of malarial infection. The present invention includes novel recombinant malarial proteins, and fragments of the same, that have unique structural and/or chemical properties that allow them to be expressed in greater quantity in heterologous systems. These recombinant proteins and fragments have immunogenic properties superior to malarial proteins expressed using different expression systems and coding sequences, but having the same primary amino acid sequence.

An artificial invasin complex is prepared from purified or recombinantly prepared invasins and gram negative bacteria lipopolysaccharides. Typically, IpaB is mixed with IpaC to form a IpaB:IpaC complex. This invasin protein complex is then mixed with the lipopolysaccharide to form an artificial invasin complex. Additional bioactive molecules can be incorporated into the complex during manufacture. This artificial invasin complex is similar in function to native Invaplex 24 or Invaplex 50. The artificial invasin complex has superior immunogenicity properties relative to the native complex and can be taylor made. Its method of preparation lends itself to scale up. The artificial invasin complex can facilitate transport of biomolecules, therapeutics and antibiotics across cell membranes in a manner similar to native Shigella Invaplex.
