    摘要: Processes and apparati are provided for separating molecules of interest from a mixture containing them which comprises subjecting the mixture to an improved method of tangential flow filtration (TFF). The improved TFF was used to clarify, and process various feedstreams for the removal of a molecule of interest. According to a preferred embodiment, a transgenic milk feedstream is stabilized and particulate matter such as fat, casein miscelles and bacteria are removed. The method of TFF used in the current invention utilizes optimized process parameters that include temperature, transmembrane pressure, cross-flow velocity, and milk concentration. Cleaning and storage procedures were also developed to ensure long membrane life. An aseptic filtration step was also developed to remove any bacteria remaining in a clarified transgenic milk feedstream.

    摘要: A system, kit and process for the separation, recovery, purification and identification of biomarkers such as peptides and the like from serum or plasma. The process is to filter the sample through a UF filtration device optionally having a vertical or substantial vertical ultrafilter membrane, especially when in a single well configuration device. The filtrate is recovered and if desired desalted using various chromatography media such as reverse phase and ion exchange media. The sample is then applied to a MALDI/TOF target or HPLC column and analyzed in a mass spectrometer the presence, absence or variation of the biomarkers. Higher recovery and concentration of these biomarkers is achieved with the present invention.

    摘要: The invention relates to a process for purifying and/or concentrating inclusion bodies (refractile bodies) in a highly concentrated particle-containing solution, in which process the inclusion body-containing suspension is directed tangentially past one or more semipermeable membranes, so that the inclusion bodies are retained behind the membranes and that substances having a relatively small molecular weight may pass through the membrane and/or be adsorbed to the membrane, and a purified and/or concentrated inclusion body suspension is obtained. In the same manner, cell wall particles are removed from a inclusion body suspension. The invention further relates to the use of a crossflow microfiltration unit for purifying and/or concentrating inclusion bodies in a suspension and for removing cell wall particles from a inclusion body preparation.
