摘要: An object of the present invention is to provide a method of identifying an amino acid site in a protein, to which a sugar is bound more accurately than in the related art, and a kit for performing the method. According to the present invention, there is provided a method for identifying an amino acid site in a protein, to which a sugar is bonded, the method including a first mass spectrometry step of subjecting a fragmented protein to mass spectrometry; for a protein in which the bonding of the sugar has been confirmed in the first mass spectrometry step, a substitution-modification step of substituting and/or modifying an amino acid of the protein at the amino acid site in the protein, to which the sugar is bonded and a fragmentation step of fragmenting the protein; and a second mass spectrometry step of subjecting the substituted and/or modified, and fragmented protein obtained by the substitution-modification step and the fragmentation step, to mass spectrometry.

摘要: The present invention relates to a high throughput targeted proteomics-based method for identifying and quantifying one or more binding molecules in a biological fluid of an individual. The present invention involves the use of a plurality of bait molecules to capture the binding molecules. The invention uses proteomics techniques, such as mass spectrometry, for the identification and quantification of the binding molecules. The methods of the invention can be used to identify and quantify antibodies produced by an individual, and proteins expressed by an individual, following exposure to antigens, such as viral antigens, particularly coronavirus antigens and more particularly SARS-CoV-2. Exposure may be via vaccination or following natural infection. The methods of the invention can be used to identify and quantify molecules which bind to a range of proteins of interest, such as autoantigens and neoantigens, as well as to viral vectors.

摘要: In a method for analyzing cells, a sample fluid having a suspending medium and cells is fed to a microfluidic device having at least one cell processing unit having a trapping region, a reaction unit, and an outlet arrangement. The trapping region is delimited by at least an input valve and a sieve valve, in particular a v-type valve that is capable of retaining the cells while letting fluids pass. The method includes trapping cells in the trapping region, subsequently establishing a flow of a reaction fluid through the trapping region while the sieve valve assumes the open state, such that the reaction fluid transfers the trapped cells from the trapping region into the reaction unit, decomposing the transferred cells into cell fragments through a decomposition process, collecting the cell fragments in the outlet arrangement, and analyzing the cell fragments.
