Abstract: Described herein are method for generating a vector for editing a cell. The method comprises ligating into a vector that encodes a portion of a gRNA a cassette comprising at least one editing cassette, a promoter, and a gene encoding another portion of the gRNA. Upon ligation, the portion of the gRNA from the editing cassette and the other portion of the gRNA are ligated and form a functional gRNA.

Abstract: This invention relates to the field of genetic engineering. Specifically, the invention relates to the construction of operons to produce biologically active agents. For example, operons may be constructed to produce agents that control the function of biochemical pathway proteins (e.g., protein phosphatases, kinases and/or proteases). Such agents may include inhibitors and modulators that may be used in studying or controlling phosphatase function associated with abnormalities in a phosphatase pathway or expression level. Fusion proteins, such as light activated protein phosphatases, may be genetically encoded and expressed as photoswitchable phosphatases. Systems are provided for use in controlling phosphatase function within living cells or in identifying small molecule inhibitors/activator/modulator molecules of protein phosphatases associated with cell signaling.

Abstract: A surgical method including positioning an optical probe within a tissue at a target location in a body of a patient; emitting electromagnetic radiation from the optical probe toward the tissue; and receiving a spectra at the optical probe; generating, based on the spectra, by a tissue classification system, a diagnosis classification indicative of a condition of the tissue and treating the tissue with a treatment modality selected based on the diagnosis classification. The spectra is generated by the tissue in response to the emitted electromagnetic radiation.

Abstract: A multi-level supply modulator for RF power amplifiers requires only a single independent voltage supply along with one or more flying capacitors to achieve multiple output levels. The flying capacitor is used to store the intermediate voltage level between the DC supply level and ground. A switch network is connected between the DC voltage supply and the power amplifier and is configured to provide power at variable, discrete DC voltages, for example the DC voltage supply level, ground, and a level halfway between the DC voltage supply level and ground. The flying capacitor is connected to switches in the switch network, and a sensing circuit detects the voltage across the flying capacitor and generates a feedback signal. Control circuitry controls the switch network (and thus the power supplied to the amplifier) based on the feedback signal.

Abstract: A slotted cannula for surgical procedures is disclosed. The slotted cannula has an elongated semi-circular body comprising a cavity extending along a longitudinal axis of the semi-circular body, the cavity having an open distal end and an open proximate end and an integrally formed outward bend extending curvilinear from the cavity on the proximate end and at least one fold line across the width of the cannula, perpendicular to the longitudinal axis. The cannula can be bent or broken along the at least one fold line to adjust the operable length of the cannula during a surgical procedure.
