Abstract: Disclosed are devices for determining an analyte concentration (e.g., glucose). The devices comprise a sensor configured to generate a signal associated with a concentration of an analyte and a sensing membrane located over the sensor. The sensing membrane comprises a biointerface layer which interfaces with a biological fluid containing the analyte to be measured. The biointerface layer comprises a biointerface polymer, wherein the biointerface polymer comprises polyurethane and/or polyurea segments and one or more zwitterionic repeating units. The biointerface layer increases sensor longevity and decrease sensor inaccuracy by inhibiting accumulation of cells, proteins, and other biological species on the outermost layers of the sensor.

Abstract: An analyte monitoring sensor configured for in vivo measurement of at least one analyte is provided, the sensor comprising a substrate having a substrate surface, an aptamer protective layer encapsulating at least a portion of the substrate surface, the aptamer protective layer permeable to the at least one analyte, one or more aptamer conjugates associated with at least a portion of the substrate surface and positioned between the aptamer protective layer and the substrate for obtaining measurements related to the at least one analyte in vivo. Methods of extending in vivo performance of the analyte monitoring sensor are also described.

The sensing membrane can also comprise an enzyme layer, wherein the enzyme layer comprises an enzyme and a polymer comprising polyurethane and/or polyurea segments and one or more zwitterionic repeating units. The sensing membrane can also comprise a diffusion-resistance layer, which can comprise a base polymer having a lowest Tg of greater than −50 C.

An adhesive can couple the base to the skin. A transcutaneous analyte measurement sensor can be coupled to the base and can be located at least partially in the host. A transmitter can be coupled to the base and can transmit analyte measurement data to a remote device.

Abstract: Devices and methods for measuring a concentration of a target analyte in a biological fluid in vivo are provided herein. In some examples, a device includes an indwelling sensor and sensor electronics. The sensor may include a substrate; a first electrode disposed on the substrate; an ionophore disposed on the substrate to selectively transport the target ion to or within the first electrode; and a second electrode disposed on the substrate. The sensor electronics is configured to generate a signal corresponding to an electromotive force which is at least partially based on a potential difference that is generated between the first electrode and the second electrode responsive to the ionophore transporting the target ion to the first electrode.
