In the present invention, there is provided a method for measuring, under an acidic pH condition, a subject substance in a specimen. The method includes: step A for forming a reaction system by mixing the specimen and an antibody against the subject substance in the specimen; and step B for measuring an antigen-antibody reaction in the reaction system, wherein a pH of the reaction system is set to be acidic, and the relationship between a pI of the antibody and the pH of the reaction system is set to be pI>pH. According to the present invention, a measurement accuracy can be enhanced in an immune measurement reaction system basically using an acidic buffer solution, particularly in a region of low concentration for an antigen.
