Abstract: In an embodiment, the present disclosure pertains to a droplet system, apparatus, or fluid sample testing system to accomplish high-precision and high-efficiency droplet manipulation (e.g., greater than 99% platform operation efficiency). In some embodiments, the droplet system, apparatus, or fluid sample testing system includes at least one microfluidic channel or chamber and at least one interdigitated electrode (IDE) that can create a localized electric field below and/or within at least one fluidic channel or chamber. In some embodiments, this allows size-specific and/or size-dependent droplet manipulation.

Abstract: In an embodiment, the present disclosure pertains to a microfluidic platform composed of a droplet generator having an entry point for donor particles and target particles, a first droplet incubation chamber in fluid communication with the droplet generator, a droplet detection functionality to allow for analysis of the inner content of droplets, and a droplet sorting functionality to allow for the separation of droplets based on the analysis of the inner content of droplets. In another embodiment, the present disclosure pertains to a method for cell-to-cell DNA, RNA, or other genetic material transfer through use of a water-in-oil emulsion microdroplet-based microfluidic platform for automation and high throughput identification or screening of genetic transfer outcomes utilizing the microfluidic platforms as disclosed herein.

Abstract: The subject invention pertains to a microfluidic apparatus and methods for screening and isolating a target cell from a population of cells. The apparatus comprises a first microfluidic layer comprising microfluidic channels; a second microfluidic layer comprising microfluidic channels; and a microfluidic cell analysis layer comprising a top hanging blocking structure located directly below each location where the first layer microfluidic channels overlap with the second layer microfluidic channels and a cell trap juxtaposed to each of the top hanging blocking structures. The top hanging blocking structures can close or open the juxtaposed cell trap when either or both the first or second layer microfluidic channels located directly above the top hanging blocking structure are sufficiently pressurized and/or sufficiently depressurized. The methods for screening and isolating a target cell from a population of cells comprise screening the population of cells using the apparatus and isolating the target cell interest therefrom.
