The present invention provides a pharmaceutical composition comprising a bispecific single chain antibody construct. Said bispecific single chain antibody construct is characterized to comprise or consist of at least two domains, whereby one of said at least two domains specifically binds to human EpCAM and comprises at least one CDR-H3 region comprising the amino acid sequence NXID antigen and a second domain binds to human CD3 antigen. The invention further provides a process for the production of the pharmaceutical composition of the invention, a method for the prevention, treatment or amelioration of a tumorous disease and the use of the disclosed bispecific single chain antibody construct and corresponding means in the prevention, treatment or amelioration of a tumorous disease.

The present invention relates to a bispecific single chain antibody molecule comprising a first binding domain consisting of one antibody variable domain capable of binding to an epitope of the human and non-chimpanzee primate CD3 epsilon chain, wherein the epitope is part of an amino acid sequence comprised in the group consisting of SEQ ID NOs. 2, 4, 6, and 8, and a second binding domain capable of binding to an epitope of a human and a non-chimpanzee primate tumor target antigen. The present invention relates to a method of identifying binding site domains that retain the capacity of binding to an epitope when positioned C-terminal of at least one further domain in a recombinant bi- or multivalent polypeptide. The present invention further relates to a kit comprising components such as panels of recombinant vectors of bacterial libraries transfected with a panel of recombinant vectors which is useful in carrying out the method of the invention. Furthermore, binding site domains and fusion proteins obtainable by the method of the invention as well as antibody-like molecules comprising such domains and proteins are described. Furthermore, pharmaceutical and diagnostic compositions containing the above-described fusion proteins and polypeptides are provided.
