摘要: Embodiments of the present disclosure are directed to methods for preparing a target polypeptide from a mixture including the target polypeptide. The method may include contacting the mixture to a hydrophobic interaction chromatography (HIC) apparatus including multiple chromatographic zones. The method may further include passing the target polypeptide through the outlets of at least a first zone and a second zone of the HIC apparatus. A residence time for the mixture including the target polypeptide in a first zone may be approximately the same as a residence time of one or more mobile phases in the second zone.

摘要: Provided herein are methods for modifying an arginine residue to provide a reactive handle suitable for attaching the arginine residue to a label, linker, or other target molecule. The methods utilize an Arnold Salt to convert the guanidine group of the arginine residue into a formylpyrimidine, and are efficient and very selective. Provided herein are mild reaction conditions for this arginine modification, making it suitable for use with complex biomolecules like proteins. The methods can include one or more additional steps to attach the modified arginine to a linker for further manipulation or for connecting the arginine residue to a target compound. Provided methods are especially useful for covalently linking an arginine-containing peptide to a target molecule such as a polynucleotide.

摘要: A method and a sensor for detecting L-arginine are provided. The method includes synthesizing ferrocene-functionalized hexadecapeptide dithiocyclopentane (FC-P16 Peptide), preparing a polypeptide composite membrane-modified electrode (FC-P16 Peptide/AuE), detecting L-Arg and other steps. The results show that the polypeptide composite membrane-modified electrode (FC-P16 Peptide/AuE) exhibits excellent electrochemical response properties to L-Arg. In 10 mmol/L phosphate-buffered saline (PBS, pH=7.4), the DPV response peak current of the polypeptide composite membrane-modified electrode has an excellent linear relationship with the L-Arg concentration of 1.0×10−13 mol/L to 1.0×10−7 mol/L, with a detection limit of 1.0×10−13 mol/L. With prominent reproducibility, repeatability and selectivity, the modified electrode has potential application in life science and nutritional health.
