Biomarkers for diagnosis and monitoring of vascular aneurysms are described in the context of the use of assays to measure a plurality of these biomarkers. Tissue degeneration, particularly elastin and/or collagen degradation, can be monitored within patient blood (serum) and/or urine to diagnose the presence, the progression, or the likelihood of rupture of aneurismal disease. Additionally, enzymes responsible for this degradation and other biomarkers responsible for the activation or inhibition of these enzymes can be monitored additionally or alternatively. Prompt diagnosis can provide the opportunity for intervention and potentially increase the health of patients by tempering the development of debilitating and life-threatening vascular aneurysms.

We hypothesized that gene expression patterns in peripheral blood cells may correlate with TAA disease status, and carried out a comprehensive gene expression survey on peripheral blood cells obtained from TAA patients and normal individuals. A distinct gene expression profile in peripheral blood cells can classify TAA patients from normal individuals. The genes provided by the present teachings define a set of diagnostic markers, thus providing a blood-based gene expression test to facilitate early detection of TAA disease. Methods of distinguishing ascending from descending TAA are also provided, as are methods of distinguishing familial from sporadic TAA.

This invention provides an isolated protein of approximately 40 kDa which is purified from human aortic tissue and immunoreactive with AAA-associated immunoglobulin. Also provided are a method of diagnosing AAA disease in a subject using said isolated protein and a pharmaceutical composition comprising said isolated protein. A method of alleviating AAA disease in a subject comprising administering said pharmaceutical composition comprising the isolated protein is also provided. The subject invention also provides a recombinantly produced human aortic protein which is immunoreactive with AAA-associated immunoglobulin. Also provided are a method of diagnosing AAA disease in a subject using said recombinantly produced protein and a pharmaceutical composition comprising said recombinantly produced protein. A method of alleviating AAA disease in a subject comprising administering said pharmaceutical composition comprising the recombinantly produced protein is also provided.
