摘要: This method for generating an indicator of the state of a patient in coma includes: generating at least one auditory stimulation by generating a sequence of auditory stimuli, the sequence producing evoked potentials in the patient; acquiring a first electroencephalographic signal produced by patient from at least one electrode; estimating at least one pair of values corresponding to a first parameter and a second parameter extracted from the first acquired signal, including estimating a first pair of values such that calculating the first parameter includes an estimation of the amplitude variance of the first signal within a predefined time window and the calculation of the second parameter includes an estimation of the correlation of two segments of the first signal; generating a state indicator for the or each pair of values of the first and second parameters, the values defining coordinates of a point in a reference base.

摘要: Plasma levels of different sub-populations of microvesicles (endothelial, leukocyte, platelet and hepatocyte) were measured by flow cytometry or ELISA/filtration on blood samples from 125 patients with cirrhosis, for which 36 of them were diagnosed with HCC at inclusion. The inventors show that the levels of microvesicles of endothelial origin (CD62E+) could predict the occurrence of HCC in patients with cirrhosis. Therefore the present invention relates to a method for determining whether a patient suffering from cirrhosis is at risk of having or developing hepatocellular carcinoma comprising determining the level of endothelial-derived microvesicles (e.g. by flow cytometry) in a blood sample obtained from the patient.

摘要: The present invention relates to a method for preparing lymphoid progenitors. The inventors took advantage of their original and relevant Warts, Hypogammaglobulinemia, Infections and Myelokathexis (WHIM) Syndrome (WS) model and the access to blood samples from five WS patients to investigate the impact of CXCR4 desensitization on BM and extra-medullary (i.e. splenic) hematopoiesis and hematopoietic stem and progenitor cells (HSPCs) recirculation. They developed, for the first time, an original in vitro system permitting to selectively expand HSPCs to obtain lymphoid progenitors by using an original cocktail of cytokines. In particular, the present invention relates to an in vitro method for preparing lymphoid progenitors by culturing HSPCs in an appropriate culture medium comprising an effective amount of a cocktail of cytokines consisting in SCF, IL-3, IL-6, IL-7, Flt-3, and CXCL12.

摘要: Acute myelogenous leukaemia (AML) often bear a mutation in the NPM1 nucleolar chaperone, but the transforming properties of the NPM1c oncoprotein remain incompletely understood. Here the inventors show that NPM1c binding to PML, a key senescence gene, disrupts PML nuclear bodies (NB) yielding proliferation, mitochondrial alterations and intracellular stress. Actinomycin-D (ActD), an anticancer antibiotic with clinical efficacy in NPM1c-AMLs, targets these dysfunctional mitochondria to induce ROS. The later disrupt disulphide-linked NPM1c/PML complex, restoring PML NBs and initiating senescence. An ActD-responsive patient displayed features of mitochondria-initiated senescence. These studies highlight unexpected mitochondrial involvement both downstream of the NPM1c/PML axis and as a key feature of ActD therapy. More particularly, the inventors pretreated AML cells with ActD and/or Venetoclax, a Bc12-targeting agent and showed that the two drugs sharply synergized to abolish clonogenic growth of NPM1c-expressing, but not control or PML-deficient cells. Collectively, these results support that combination of antineoplastic antibiotics and BCL-2 inhibitors would be suitable for the treatment of NPM-1-driven acute myeloid leukemia (AML).

摘要: The present invention relates to tetramolecular parallel G-quadruplex-forming oligonucleotides. If G-quadruplexes are of prime importance in biology, their use is hampered by the propensity of G4-prone DNA molecules, in particular G4-prone DNA molecules of long size, to adopt many different G4 topological conformations or other alternative foldings. By introducing a lipid modification at the end of the oligonucleotide, the inventors succeeded in obtaining long tetramolecular parallel G-quadruplexes (tpG4). The present invention thus concerns an oligonucleotide modified by substitution at the 5′ or the 3′ end by a lipid moiety, wherein said oligonucleotide comprises a nucleic acid sequence of at least 10 nucleotides, said nucleic acid sequence including a series of at least 4 consecutive guanine residues located in the middle of said sequence. A tetramolecular parallel G-quadruplex comprising 4 identical modified oligonucleotides as defined above, wherein each of the 4 consecutive guanine residues included in the middle of the nucleic acid sequence of each oligonucleotide respectively form G-quartets with the corresponding guanine residues of the other 3 oligonucleotides, said G-quartets being stabilized by π-π staking and Hoogsteen hydrogen bonding, is also contemplated. The modified oligonucleotides have preferably the general formula (I) or (II), wherein the oligonucleotides are modified by substitution at the 5′ or the 3′ end by a lipid moiety, and said oligonucleotides comprise a nucleic acid sequence of at least 10 nucleotides, said nucleic acid sequence including a series of at least 4 consecutive guanine residues located in the middle of said sequence.
