摘要: An improved system for large scale production of proteins and/or polypeptides in cell culture, particularly in media characterized by one or more of: i) a cumulative amino acid concentration greater than about 70 mM; ii) a molar cumulative glutamine to cumulative asparagine ratio of less than about 2; iii) a molar cumulative glutamine to cumulative total amino acid ratio of less than about 0.2; iv) a molar cumulative inorganic ion to cumulative total amino acid ratio between about 0.4 to 1; or v) a combined cumulative glutamine and cumulative asparagine concentration between about 16 and 36 mM, is provided. The use of such a system allows high levels of protein production and lessens accumulation of certain undesirable factors such as ammonium and/or lactate. Additionally, culture methods including a temperature shift, typically including a decrease in temperature when the culture has reached about 20-80% of it maximal cell density, are provided. Alternatively or additionally, the present invention provides methods such that, after reaching a peak, lactate and/or ammonium levels in the culture decrease over time.

摘要: A power-retractable feeder for an injection molding machine in which an actuator is adapted to forcibly seal a button-type outlet nozzle of the feeder against a side inlet port of a charging cylinder without developing an unbalanced lateral load on the cylinder. As disclosed, the reaction force developed on the actuator is transmitted to the side of the cylinder opposite the inlet. The actuator is selectively operable to retract the feeder nozzle from the cylinder for inspection, removal, maintenance, or the like. The disclosed feeder is of the screw type driven by a rotary motor through a coupler which isolates an axial reaction force on the screw from the motor. Also disclosed is a remote actuator and associated linkage for operating a shut-off valve in the feeder nozzle at either the extended or retracted position of the nozzle.

摘要: Mixing means in the form of a torpedo for an injection molding machine having a plurality of alternately isolated and intersecting flow paths for repeatedly dividing and recombining material flow streams to achieve uniform properties throughout the material. The paths are formed as right and left-hand helical channels on the circumference of the torpedo body, with the helixes of one hand having a pitch different from that of the other such that the flow paths intersect in a pattern which is bilaterally asymmetrical with respect to an axial net flow direction. The nonsymmetrical arrangement is formed to provide substantially greater mixing action than that obtainable with similar symmetrical arrangements.

摘要: A thermosetting injection molding machine includes an injector chamber, a distribution platen, four distribution nozzles, and a mold heating platen. A thermosetting material flows from the injector chamber to two crossed distribution chambers each of which extends from edge to edge completely through the distribution platen. Liquid coolant passages extend through the distribution platen along both sides of each of the distribution chambers. The distribution nozzles include cores which block the ends of the distribution chambers and which carry the thermosetting material through the mold heating platen to the mold. The distribution nozzles also include jackets having a greater coefficient of heat conductivity than the cores for maintaining all but the tip of the cores adjacent the mold at a temperature below the curing temperature of the thermosetting material. A removable mold for the machine includes a mold cluster adjacent each of the four distribution nozzles. The mold clusters each include a plurality of mold cavities and a runner for carrying the thermosetting material from its associated distribution nozzle to its associated mold cavities. By this arrangement, a multiple nozzle distribution system and a mold heating system are provided by the machine completely independently of the mold to simplify mold design.
