    摘要: Described is a novel family of cell surface serpentine transmembrane antigens. Two of the proteins in this family are exclusively or predominantly expressed in the prostate, as well as in prostate cancer, and thus members of this family have been termed “STEAP” (Six Transmembrane Epithelial Antigens of the Prostate). Four particular human STEAPs are described and characterized herein. The human STEAPs exhibit a high degree of structural conservation among them but show no significant structural homology to any known human proteins. The prototype member of the STEAP family, STEAP-1, appears to be a type IIIa membrane protein expressed predominantly in prostate cells in normal human tissues. Structurally, STEAP-1 is a 339 amino acid protein characterized by a molecular topology of six transmembrane domains and intracellular N- and C-termini, suggesting that it folds in a “serpentine” manner into three extracellular and two intracellular loops. STEAP-1 protein expression is maintained at high levels across various stages of prostate cancer. Moreover, STEAP-1 is highly over-expressed in certain other human cancers.

    摘要: Compositions for the diagnosis and therapy of prostate and colon cancer, derived from or based on a novel prostate-specific, androgen-related, cell membrane associated and secreted serine protease termed 20P1F12/TMPRSS2 are described. A full length cDNA comprising the entire coding sequence of the 20P1F12/TMPRSS2 gene (also designated 20P1F12-GTC1 herein) is provided (FIG. 1). Among the compositions provided are antibodies that bind to 20P1F12/TMPRSS2 proteins and polypeptide fragments thereof, including antibodies labeled with a detectable marker or toxin or therapeutic composition. The invention also provides prognostic and diagnostic methods of examining a biological sample for evidence of disregulated cellular growth by comparing the status of 20P1F12/TMPRSS2 in the biological sample to the status of 20P1F12/TMPRSS2 in a corresponding normal sample, wherein alterations in the status of 20P1F12/TMPRSS2 in the biological sample are associated with disregulated cellular growth. The invention further provides various therapeutic compositions and strategies for treating prostate cancer, including particularly, 20P1F12/TMPRSS2 polypeptide and anti-20P1F12/TMPRSS2 antibody therapy methods and compositions, cancer vaccines, and small molecule therapy.

    摘要: The screening efficiency of a gyratory screening machine, of the type having a moving base, is significantly increased by mounting to the base a spring/mass system which is tuned so that operation of the machine drive produces amplified movement of the mass along at least one of two perpendicular axes on which the base moves. Movement of the mass reduces the net force on the base along the axis on which the mass oscillates. The reaction force of the machine drive on the base along the other of the two perpendicular axes may be offset by a rotary counterbalance operated by the drive. Movement of the base relative to the ground is thereby reduced, and movement of the screen deck relative to ground is increased. This both increases the effectiveness of the screening motion and decreases the total reaction force transmitted through the base.

    摘要: A screening machine of the type used to separate or classify mixtures of solid particles of different sizes includes a fixed base and a perforate screen deck mounted for movement relative to the base during a screening operation. Leaf springs connect the tail end of the screen deck to the fixed base so that the leaf springs support the tail end of screen deck for movement relative to the base. The leaf springs are oriented horizontally to avoid introducing vertical movement to the screen deck as it moves to and from the head and tail ends. The leaf springs are offset or deflected when the machine is at rest or in the neutral mid-point of its stroke. This arrangement advantageously utilizes the bias of the leaf springs to accelerate, assist or enhance movement of the screen deck during portions of its stroke and likewise to decelerate, hinder or retard the movement during other portions of its stroke. With an appropriate offset, an increase in the effectiveness of the screening process (i.e., fewer defective particles passing through the screen deck) while a decrease in the screening time is attained.

    摘要: A coupling for directing a stream of particles falling from a feed spout through an opening on the top cover of the screen box of a screening machine. An upper diaphragm seals the feed spout within the mouth of a funnel. A lower diaphragm seals the funnel discharge opening within the top cover opening. Both diaphragms are flexible and permit the funnel to tilt axially and accommodate movement of the top cover opening with respect to the feed spout. Optionally, a gimble may support the funnel and diaphragms and thereby relieve the diaphragms from the weight of the coupling.

    摘要: A stiffened composite is provided exhibiting multi-directional reinforcement with continuous long fibers. The composite has a substrate and serpentine stiffeners adhered thereto. The stiffeners merge toward, join with and diverge from adjacent stiffeners to provide a stiffener matrix on the substrate. The serpentine stiffener permit the matrix to exhibit multi-directional reinforcement with the continuous long fibers within the serpentine ribs. The composite structures are useful as aircraft bypass ducts, and as other composite products.

The weight of the force reducer, the spring constant of the spring which oscillates the force reducer, and overall cost are surprisingly reduced.
