    Abstract: A perfusion bioprocessing system (10) includes a bioreactor (12) and a recirculation flow path (14) provided with at least in one first feed flow control device (46) and at least one second feed control device (48). The perfusion bioprocessing system (10) further includes a first tangential flow filter (16) coupled to the bioreactor (12) via the recirculation flow path (14) and a second tangential flow filter (18) coupled to the bioreactor (12) via the recirculation flow path (14). The first tangential flow filter and the second tangential flow filter are coupled to a permeate flow path and a retentate flow path. Additionally, the perfusion bioprocessing system (10) includes a control unit (90) coupled to the at least one first feed flow control device (38) and the at least one second feed control device (40).

    Abstract: Systems (700) and methods (800) for method of continuous fluid flow in a bioreactor (700) is provided. The method (800) comprises providing (805) a bioreactor system (700) including a bioreactor volume (720), a filtration part (730), and a recirculation line (721) including a recirculation pump (722) is provided between the bioreactor volume (720) and the filtration part (730). The method (800) further comprises providing (810) a plurality of sensors (760) along the recirculation line (721) and monitoring the fluid flow parameters using the sensors (760). The method further comprises sending (820) a plurality of signals from the sensors (760) indicative of the fluid flow parameters to one or more controllers; and controlling (830) the fluid flow rate at the recirculation pump (722) by means of the or each controller.

    Abstract: The present subject matter discloses a bioreactor apparatus. The bioreactor apparatus comprises a bioreactor vessel configured to culture cells. The bioreactor apparatus furthermore comprises a sensor configured to measure Dissolved Oxygen (DO) in the bioreactor vessel. The DO measurements comprise a plurality of DO values recorded at, at least, a plurality of time instances during operation of the bioreactor apparatus. The bioreactor apparatus furthermore comprises a controller configured to obtain the DO measurements. The controller furthermore is to determine, in real-time or approximately real-time, an oxygen mass transfer co-efficient (kLa) associated with the operation of the bioreactor apparatus. Furthermore, the controller is configured to control, in real-time or approximately real-time, at least one cell culture parameter associated with the operation of the bioreactor apparatus based on the kLa.
