    摘要: A machine or apparatus simultaneously manufacutures a core-reinforced welded panel and assembles core strips in operative relationship with the face sheets of said panel. The machine includes a first welding head which is insertable in the space between the face sheets and has welding electrode means thereupon which internally weld securement portions of the core strips to adjacent inner surfaces of face sheets of any desired thickness. The welding head includes movable jaws mounting the welding electrodes to move the welding electrodes into engagement with the securement portions of the core strips and the welding electrodes are resilient to permit them to move independently of the movable jaws and each electrode provides an electrically isolated weld. A second welding head is provided which is capable of securing web portions of the core strips by weldments to one another after the weldment of the core strips at their securement portions has been accomplished by the first welding head. Various adjustments and indexing mechanisms permit the machine to fabricate panels of different dimensions and different size core without the necessity for the extensive down-time necessitated by the utilization of prior art machines.

    摘要: A panel includes metallic face sheets maintained in operative relationship with each other by a metallic core structure. The core structure consists of a plurality of securement core strips which may be internested or juxtaposed to provide a honeycomb core configuration. Structural integrity is created by the provision of internally generated welds which fasten securement portions of the core strips in operative relationship with the face sheets, but which can be terminated inwardly of the outer surfaces of the face sheets resulting in a very small volume of face sheet material being cast into a weld nugget and being heat affected and maintaining the integrity of the face sheets while, nevertheless, providing maximum structural strength and eliminating face sheet thickness limitations.Also disclosed is a method of fabricating the metallic panel which includes the steps of internally generating the weldments so that, by minimum pressure and minimum energy, discrete welds are formed which fasten the securement portions of the core strips in operative relationship with the face sheets while greatly reducing the dimensions of the weld nuggets in comparison with the nuggets which result from the utilization of prior art external welding techniques.

    摘要: Wild-type bacteria with polyurethanase activity, and the ability to excrete polyurethanase into a culture medium have been genetically altered to significantly increase their polyurethanase activity. Biologically pure cultures of these genetically altered are cultured in a medium containing an inducer for the production of polyurethanase. The bacteria are then removed from the culture medium to provide a preparation with a high polyurethanase activity. One useful genetically altered bacteria is a strain of Pseudomonas chlororaphis designated ATTC No. 55729. The polyurethanase-containing preparation may then be used directly to degrade polyurethane, particularly polyurethane and polyurethane-based coatings.

    摘要: Biological toxins are indirectly detected by using polymerase chain reaction to amplify unique nucleic acid sequences coding for the toxins or enzymes unique to toxin synthesis. Buffer, primers coding for the unique nucleic acid sequences and an amplifying enzyme are added to a sample suspected of containing the toxin. The mixture is then cycled thermally to exponentially amplify any of these unique nucleic acid sequences present in the sample. The amplified sequences can be detected by various means, including fluorescence. Detection of the amplified sequences is indicative of the presence of toxin in the original sample. By using more than one set of labeled primers, the method can be used to simultaneously detect several toxins in a sample.

    摘要: The fluorescent detection of target DNA is enhanced by amplifying the DNA in a sample by initiating a polymerase chain reaction using a first pair of primers and an appropriate polymerase. The DNA is then reamplified, this time using a second pair of nested primers which bracket DNA base sequences within the region bracketed by the first set of primers. At least one of the second pair of primers is bound to a fluorescent reporter molecule and at least one of the second pair of primers has bound thereto a specific binding site for a double-stranded DNA-binding protein. The thus amplified sample is then contacted with a fluorescent sensor having the appropriate double-stranded DNA-binding protein attached thereto. Then, the fluorescent reporter molecule is excited by light of the appropriate wavelength. The present invention provides greater sensitivity, adaptability and ease of use than previously available.
