Abstract:
The invention encompasses cell-based systems comprising biomarkers that respond to oxidative stress (OS) in a quantitative manner, and methods of use thereof. The systems are useful for screening, identifying and testing antioxidant agents, combinations, and formulations thereof for preventing, treating, or reducing symptoms of conditions associated with oxidative damage to cells. The cell-based systems are useful for identifying effective new antioxidant agents and for optimizing antioxidant formulations for targeted therapeutic applications. One cell-based system utilizes RPE cells of the eye for identifying and optimizing antioxidant compositions effective for treatment of age-related conditions such as macular degeneration. The cell-based systems provide a convenient, inexpensive and physiologically relevant in vitro alternative to human population-based methods for testing efficacy of nutritional and pharmaceutical compositions comprising antioxidants. The invention further encompasses nutritional or pharmaceutical compositions targeting particular diseases such as AMD, formulated using methods as disclosed herein.

Some embodiments of the invention provide a self-priming adapter system including a pump inlet receiving liquid mixed with air, a pump coupled to the pump inlet, and a self-priming adapter coupled to the pump. The self-priming adapter includes a separation chamber. The separation chamber includes one or more baffles to substantially separate the air from the liquid. The self-priming adapter system also includes a recirculation line coupled to the self-priming adapter to return the liquid to the pump inlet. Some embodiments of the invention provide a self-priming adapter for use with existing pumps of any type. Some embodiments of the invention provide a method of self-priming a pump using a self-priming adapter.

The present disclosure provides novel hemi- and mono-salts of 7-[(3R,4R)-3-Hydroxy-4-hydroxymethyl-pyrrolidin-1-ylmethyl]-3,5-dihydro-pyrrolo[3,2-d]pyrimidin-4-one (Compound 1) with various organic and inorganic acids. In one embodiment, the organic acid is a C4 organic diacids. The present disclosure further provides novel methods for preparing these salts. The novel monohydrates hemisalts of the C4 organic diacids are isostructural and can be prepared with different properties. Multiple acids can be used simultaneously and the proportion of acids can be varied offering the opportunity to select hemi-salts of compound 1 with desired properties.

The present invention provides improved dietary supplements and methods for inhibiting the progression of macular degeneration and promoting healthy vision. The dietary supplements of the invention contain cobeadlets comprising vitamin E and carotenoids in the form of Vitamin A, lutein and/or zeaxanthing. The dietary supplements of the invention further contain Vitamin C, copper and zinc and may also contain such ingredients as rosemary, DHA, other vitamins and minerals.
