    摘要: Muteins of enzyme acceptor polypeptide fragments of .beta.-galactosidase are provided which exhibit substantially increased kinetic complementation activity with no significant loss in stability. A preferred enzyme acceptor fragment has an amino acid other than cysteine located at position 500 of the natural sequence. An especially preferred substitution is serine or valine. Other preferred muteins have an amino acid other than methionine located at position 443, with leucine being especially preferred, or an amino acid other than cysteine at position 76, with leucine being an especially preferred substitution. Also provided are methods for producing the novel muteins, reagent compositions comprising the novel muteins, and immunoassay methods for determining an analyte in which the novel mutein recombines with an enzyme donor polypeptide fragment to form enzymatically active .beta.-galactosidase.

    摘要: This invention is directed to a lateral flow assay for detecting the presence of an analyte in a liquid test sample. The lateral flow assay represents an improvement in the ability to accurately and with high fidelity to detect the presence or absence of a target analyte in a liquid sample, in part, by encompassing a reference region of immobilized, non-diffusible analyte that allows for detection of any factors that interfere with the interaction and binding of the analyte to the labeled capture reagent. Any influences on the interaction and binding of the analyte that is free in solution in the liquid test sample to its complementary labeled reagent will be encountered in parallel in the binding between the immobilized analyte in reference region to the labeled reagent as it diffuses through the reference region. In one embodiment, the lateral flow assay of the invention is a urine-based human Chorionic Gonadotropin (hCG) assay.

    摘要: A readhead for illuminating a sample carrier and receiving light from the sample carrier, including a housing for receiving a sample carrier, an array of light sources mounted within the housing in a fixed position relative to the sample carrier, and including first and second light-emitting diodes for emitting substantially monochromatic light of a two different wavelengths, a light guide mounted in the housing between the light-emitting diodes and the sample carrier, and a light detector coupled to receive light from the sample carrier. The readhead also includes a light source for directing excitation light of a predetermined wavelength to the sample carrier, and a light filter positioned between the sample carrier and the light detector and adapted to prevent passage therethrough of the excitation light. The readhead allows both fluorescence spectroscopy and reflectance spectroscopy to be conducted on the sample carrier.

    摘要: A specific binding assay, e.g., immunoassay, method and reagent system wherein a structural analog of the label portion of the labeled reagent is included in the reaction mixture to interact with potentially interfering substances in the test sample. The label analog is selected or designed to be substantially inactive in the assay detection system. The invention improves the precision of the assays, particularly homogeneous immunoassays, by significantly reducing or eliminating the variable interaction, e.g., binding, of the labeled reagent by interfering substances in the test sample.
