    摘要: The present invention provides novel compositions of binding moiety-nanoparticle conjugates, aggregates of these conjugates, and novel methods of using these conjugates, and aggregates. The nanoparticles in these conjugates can be magnetic metal oxides, either monodisperse or polydisperse. Binding moieties can be, e.g., oligonucleotides, polypeptides, or polysaccharides. Oligonucleotide sequences are linked to either non-polymer surface functionalized metal oxides or with functionalized polymers associated with the metal oxides. The novel compositions can be used in assays for detecting target molecules, such as nucleic acids and proteins, in vitro or as magnetic resonance (MR) contrast agents to detect target molecules in living organisms.

    摘要: The invention provides a method of identifying an antigen that is present in different amounts in two different samples. In general, the methods involve generating a first and second distinguishably labeled population of antibodies that reactive to the two samples, contacting the first and second labeled populations of antibodies with a plurality of antigens; and identifying any resultant antigens that are differentially bound by the first and second populations of antibodies. The antigens may be on the surface of cells e.g., animal cells, or on a solid support. Once identified, the nucleic acid encoding an antigen of interest may be identified and sequenced to reveal the identity of the antigen of interest. Kits for performing the methods are also provided. The methods find most use in medical and research applications, in particular, for identifying cell surface targets for immunotherapy and drug discovery.

    摘要: A method of analyzing a sample for one or more analytes of interest includes: providing an element having a base layer; a layer containing streptavidin; a spreading layer, wherein the streptavidin may or may not be in the spreading layer; providing immunocomponents and labeled immunoreactants which may be in the spreading layer or may be combined with the sample; dispensing the sample, and optionally the immunocomponents and a labeled immunoreactant onto the spreading layer at three or more areas, wherein each center of the three or more areas is substantially equidistance to the center formed by the three or more areas, and wherein each area contacts an adjacent area such that wash fluid flow in any direction will contact sample; washing the sample and label by directing the wash fluid at the center formed by the three or more areas, whereby the wash fluid equally flows over each of the three or more areas; and taking a measurement at each three or more areas to determine the presence or concentration of the one or more analyte. In a preferred embodiment, the element is a dry slide used on a diagnostic analyzer.

    摘要: This invention is directed to a lateral flow assay for detecting the presence of an analyte in a liquid test sample. The lateral flow assay represents an improvement in the ability to accurately and with high fidelity to detect the presence or absence of a target analyte in a liquid sample, in part, by encompassing a reference region of immobilized, non-diffusible analyte that allows for detection of any factors that interfere with the interaction and binding of the analyte to the labeled capture reagent. Any influences on the interaction and binding of the analyte that is free in solution in the liquid test sample to its complementary labeled reagent will be encountered in parallel in the binding between the immobilized analyte in reference region to the labeled reagent as it diffuses through the reference region. In one embodiment, the lateral flow assay of the invention is a urine-based human Chorionic Gonadotropin (hCG) assay.

    摘要: A polysaccharide coated carrier having a coating of at least two successive layers of polysaccharide. The first polysaccharide layer spontaneously associates with a second polysaccharide layer and, optionally, the carrier. Each successive layer of polysaccharide spontaneously associates with a preceding layer. Spontaneous association occurs due to the presence of oppositely charged functional groups on each layer of polysaccharide or due to a spontaneous reaction between the functional groups the layers. The carrier may be any surface such as a tube, microtitration plate, bead, particle or the like and is suitable for use in diagnostic or therapeutic methods.
