This invention is a tool for plunge cutting comprising a motor, a transmission connected to the motor and a blade assembly connected to the transmission, wherein the blades move in an oppositely reciprocating and sliding manner to provide plunge cuts of variable depth with closed ends and a continuous perimeter. The invention is designed to be held in juxtaposition to the area to be cut, and the invention further optionally includes a novel blade wherein the blades have opposite first and second sides, wherein the first side is completely flat, allowing it to slide unobstructed against the first side of the other blade; and wherein alternating teeth, including both end teeth, are straight and not bent or set; and wherein alternating even teeth are bent toward the second side of the blade; and wherein one-half of the teeth point forward and the other half of the teeth point aft; and wherein the blade teeth follow an arc; and wherein the front and aft edges of the blade are shaped to form a gullet.

The pharmacophore model may further comprise a heteroatom in the first plane A. In some embodiments, the distance between the center of the first plane A and the center of the second plane B is about 6.5 to about 9.5 Å. In some embodiments, the distance between the center of the first hydrophobic moiety C and the center of the second hydrophobic moiety D is about 8.0 to about 16.0 Å. In some embodiments, the distance between the center of the first plane to the center of the first hydrophobic moiety C is about 3.0 to about 5.0 Å. In some embodiments, the distance between the center of the second plane to the center of the second hydrophobic moiety C is about 3.0 to about 5.0 Å. In this application is described substrates for high-throughput assays of clostridial neurotoxin proteolytic activities. Two types of substrates are described for use in assays for the proteolytic activities of clostridial neurotoxins: (1) modified peptides or proteins that can serve as FRET substrates and (2) modified peptides or proteins that can serve as immobilized substrates. In both types a fluorescent molecules is present in the substrate, eliminating the requirement for the addition of a fluorigenic reagent. The assays described can be readily adapted for use in automated or robotic systems.

Substrates of botulinum toxin serotype A (BoNT A), kits comprising the substrates, and methods of using the substrates are disclosed. BoNT A cleaves SNAP-25 and the substrates that are based upon a portion of SNAP-25. Various amino acid modifications to the sequence of the peptide based on SNAP-25 are performed. Fluorescence labels that act as donors and acceptors may be added to the substrate to aid in the study of BoNT A.
