    摘要: Provided is a method for producing viral genomic and complementary DNA expression libraries and novel viral polypeptides without requiring virus cultivation prior to library construction. The method includes direct isolation of viral particles from the environment by differential filtration and centrifugation. The viral nucleic acids are extracted and used to construct libraries that may be screened by one of several methods to identify useful coding sequences or may be used to produce novel polypeptides.

The method comprises the steps of: (a) separately microencapsulating in a semi-permeable membrane each or a small number of microorganisms from a microorganism population containing said mutant; (b) growing said microencapsulated microorganisms including treating to induce a detectable difference between microcapsules containing mutant microorganisms and those containing non-mutant microorganisms; and (c) separating said microcapsules containing mutant microorganisms from those containing non-mutant microorganisms based on said difference.

    摘要: Methods and systems for purifying cells and/or viruses are provided. The sample is added to a well disposed in a medium. A potential is applied across the medium to cause the contaminants to enter one or more walls of the well, and retain the cells and/or viruses in the well. The cells and/or viruses can be removed from the well, and optionally adhered or fixed to a surface, or detected. In one embodiment, the cells and/or viruses may be retained in the well by embedding in the medium. The medium including the embedded cells and/or viruses may be excised or otherwise removed and transferred to a glass slide or other solid surface.
